Synthesis of certain tricyclo[5.2.1.02,6]decane derivatives of antihypertensive potential

A series of tricyclo[5.2.1.02,6] decane derivatives 1a-d, 2,3 and 20a-c related to the antihypertensive drugs Indoramine and Guanethidine were synthesized. The target compounds were prepared through oximation of the tricyclic ketone 8 and subsequent reduction to the endo amine analogues 5, 10, 13. Reaction of the amines with various reagents depicted in Scheme1, 2, 3 lead to the required products. The desired compounds were screened for their antihypertensive activity in desoxycorticosterone acetate (DOCA) / salt-hypertensive rats by using hydralazine as reference drag. All the compounds showed antihypertensive activity where 1c and 20c were the most active in the series.
