Cyclization synthesis method of naphthalene nitrile and derivatives thereof

The invention relates to a cyclization synthesis method of naphthalene nitrile and derivatives thereof. The method comprises the following steps: stirring benzonitrile ketene, a fluorine-containing substance, a benzyne precursor and anhydrous acetonitrile at room temperature under an oxygen isolation condition until the reaction is finished; distilling the reaction solution under reduced pressureto obtain a solid substance; and carrying out silica gel column chromatography separation and purifying on the solid substance to obtain the naphthalene nitrile and the derivatives thereof. The cyclization synthesis method of the naphthalene nitrile and the derivatives thereof is simple and convenient to operate, environment-friendly, high in yield, wide in substrate application range, free of transition metal catalysis and economical in process.

A mild and transition-metal-free benzannulation reaction for the construction of the naphthalene skeleton has been described. Benzynes react with α-cyano-β-alkylenones through a tandem nucleophilic addition/cyclization/aromatization process to afford polysubstituted naphthalenes in 50-94% yields.
