A novel series of diphenylbutylpiperidines as autophagy inducers was described and extensive SAR studies resulted in derivatives (15d-e, 15i-j) with 10-fold greater activity than the lead compounds 1 and 2. Meanwhile, a new synthetic route to diphenylbutyl bromide (6) from bromobenzene and γ-butyrolactone was also reported here.

1--4-(4-chloro-3-trifluoromethylphenyl)-4-piperidinol (I) was transformed by reactions with acetyl chloride, decanoyl chloride and 3,4,5-trimethoxybenzoyl chloride to the esters II-IV.The acetate II, administered orally, was compared with the amino alcohol I in the tests of antiapomorphine actions in rats and dogs and was found inactive.The decanoate III, which was administered intramuscularly to dogs, proved properties of an ultra-long-acting depot neuroleptic.
