DPN I is sensitive to methylation, and its activity is inhibited by the presence of 5-methylcytosine at the indicated site, provided that no 6-methyladenine is present.

Uses

Used in PCR-mediated mutagenesis:
DPN I is used as a digestion enzyme for wild type DNA after PCR. This application is crucial for the process of site-directed mutagenesis, where the wild type DNA template is digested, allowing for the incorporation of mutated sequences.
Used in Ligation and Recutting Assays:
In a typical ligation and recutting assay, DPN I fragments obtained from the complete digestion of 1μg pBR322 DNA are ligated for 16 hours at +4°C with 1U T4 DNA Ligase in a buffer containing 66mM Tris-HCl, 5mM MgCl2, 5mM Dithiothreitol, and 1mM ATP, pH 7.5 (at +20°C). This process allows for the assessment of the percentages of product that can be ligated and subsequently recut with DPN I, yielding the typical pattern of pBR322 × Dpn I fragments.
