Method for reducing amide compound into amine compound under cobalt catalysis

The invention discloses a method for reducing an amide compound into an amine compound by cobalt catalysis, which comprises the following steps: under the protection of inert gas, mixing a raw material amide compound (II), a reducing agent silane, an activating agent sodium triethylborohydride, a catalyst aminopyridine imine tridentate nitrogen ligand cobalt complex (I) and a reaction solvent, reacting at 100-130 DEG C for 6-24 hours, and carrying out post-treatment on the reaction liquid to obtain a product amine compound (III). The invention provides an effective method for preparing the amine compound by catalyzing the amide reduction reaction through cobalt by utilizing the amine pyridine imine tridentate nitrogen ligand cobalt complex catalyst, and compared with the existing method, the method is simple to operate, mild in reaction condition, wide in reaction substrate application range, high in selectivity, stable in catalyst and high in efficiency; the method has a relatively high practical application value in synthesis;.

Synthesizing aminesviathe hydroaminomethylation (HAM) reaction of olefins, a multicomponent reaction, has been regarded as one of the most attractive methods compared with the traditional methods considering the atom economy and environmental friendliness. However, the use of homogeneous catalysts, complex ligands containing diphosphine or nitrogen, and base or acid additives has severely hampered the utilization of these methods. Herein, an efficient heterogeneous Ru/TiO2-catalyzed HAM reaction of olefins is developed without any additives. Various amines, including secondary and tertiary amines, can be successfully obtained from olefins including aromatic and aliphatic olefins. Systematic studies demonstrate the lower electron density of Ruδ+and the higher number of acid sites of Ru/TiO2, leading to the high HAM reaction activity of olefins. Most importantly, nitrobenzene derivatives can also be transformed to the corresponding products over Ru/TiO2in excellent yields.

Herein, a novel route to synthesize unsymmetrically N,N-disubstituted formamides is reported, which is achieved via reductive coupling of primary amine and aldehyde with CO2/H2 over a cobalt-based catalytic system composed of CoF2, P(CH2CH2PPh2)3 and K2CO3. The mechanism investigation indicates that a secondary amine is formed via hydrogenation of the imine originated from aldehyde and primary amine, which further reacts with HCOOH generated from CO2 hydrogenation, resulting in the formation of NNFA finally.

The excitation of a RuII photosensitizer in the presence of ascorbic acid leads to the reduction of iminium ions to electron-rich α-aminoalkyl radical intermediates, which are rapidly converted into reductive amination products by thiol-mediated hydrogen atom transfer (HAT). As a result, the reductive amination of carbonyl compounds with amines by photoredox catalysis proceeds in good to excellent yields and with broad substrate scope and good functional group tolerance. The three key features of this work are 1) the rapid interception of electron-rich α-aminoalkyl radical intermediates by polarity-matched HAT in a photoredox reaction, 2) the method of reductive amination by photoredox catalysis itself, and 3) the application of this new method for temporally and spatially controlled reactions on a solid support, as demonstrated by the attachment of a fluorescent dye on an activated cellulose support by photoredox-catalyzed reductive amination.
