Synthesis and antihypertensive activity of 3-amino-4-(arylideneamino)-4H-1,2,4-triazoles

A series of 3-amino-4-(arylideneamino)-4H-1,2,4-triazoles was synthesized with the aim to find new antihypertensive compounds. The hypotensive activity was examined orally in conscious spontaneously hypertensive rats and compared with that of clonidine, guanfacine, nebidrazine and hydralazine. Six compounds were chosen for dose-response studies. The compounds 3-amino-4-(2,6-dichlorobenzylideneamino)-4H-1,2,4-triazole and 3-amino-4-(2,6-dimethylbenzylideneamino)-4H-1,2,4-triazole showed highest activity, but did not possess higher potency than the reference substances.
