The invention discloses a hyperbranched polysiloxane-based blocking agent and a preparation method thereof. The method comprises the steps that erythritol serves as a raw material, the erythritol is sequentially subjected to an oxidation reaction, an amination reaction and a Michael addition reaction in a solution, and therefore a final product is obtained. The method is simple in process, low incost, mild in condition, good in safety, high in yield and suitable for industrial production; the prepared hyperbranched polysiloxane-based blocking agent has the characteristics that the molecular size and structure are accurate and controllable, the end group contains siloxane groups, and the group density is high, can be quickly gelled and deposited to form a compact blocking layer after entering a reservoir seepage channel, and effectively blocks drilling fluid filtrate from invading the seepage channel, so that the well wall stability is greatly improved, the water-based drilling fluid prepared by taking the agent as a key auxiliary agent can effectively inhibit hydration expansion and dispersion of bentonite and improve the rolling recovery rate of shale.
