DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) In dogs treated orally with phenobarbital [50-06-6], diphenylhydantoin [630-93-3], primidone [125-33-7], or polybrominated biphenyls, only phenobarbital increased serum alk. phosphatase [9001-78-9]. No histol. changes were obsd. in the liver of any of the treated dogs. Cefazolin [25953-19-9] was given s.c. to rats at 2 g/kg/day on days 1-18 and at 0.5 g/kg/day on days 19-32. Isoniazid [54-85-3] was given orally at 20 mg/kg/day for 33 days to a similar group. Half of each group received pyridoxal-HCl [65-22-5], s.c., at 0.2 mg/kg/day on days 1-18 and at 2 mg/kg/day on days 19-46. Serum alanine aminotransferase [9000-86-6] decreased in each group, but returned to normal on day 32 in rats dosed with the isoniazid-pyridoxal combination. Isoniazid is known to interfere with alanine aminotransferase activity by decreasing the availability of the coenzyme. The mechanism of cefazolin has not been elucidated.

The blood sample is extd. with CH2Cl2 and sepd. on a glass column packed with 3% OV-17 on Chromosorb W HP using N as the carrier gas. A recovery of 98-100% was obsd. The limit of detection is approx. 0.5 mg/mL. The procedure is highly reproducible with variations from 0.5-1.3 mg/mL. Kemadrin and Prolixin did not interfere with the detn. of I, II, III, and IV.
