DOCUMENT TYPE: Report CA Section: 4 (Toxicology) Cycloheximide [66-81-9] when added to the proliferating splenocyte system at 0.005 mg/mL caused restriction of cells in the late G1 phase of the cell cycle and delayed the passage of cells into the S and G2 + M phases. Cycloheximide was chosen as a pos. control. Mitomycin C [50-07-7], a cell blocking agent, produced inconsistent results using the splenocyte system and was ruled out as a pos. control. Butylated hydroxytoluene (BHT) [128-37-0], at a threshold dose of 20 mg/mL in the splenocyte system, enhanced the G1 phase and shortened the cell cycle in agreement with published reports. Phenylpropanolamine [14838-15-4], at a threshold dose of 100 mg/mL in the splenocyte system, caused a complete depression of the cell cycle with little or no indication of cellular toxicity. Anal. of this data detd. a minimal ED for each compd. thereby meeting the exptl. goal of quantitation of cell cycle perturbation.
