The enzyme was calibrated using citric acid-Cu ion buffers, and the reaction was 50% inhibited at a pCu of 8.0. Metal ion buffers prepd. from citric acid [77-92-9], ethylenediamine [107-15-3], bicine [150-25-4], and albumin ligands were used to check the accuracy of the measurements of free Cu by ion-selective electrodes. The inhibition profiles of the enzyme for the 4 metal ion buffers gave a 50% inhibition value of 7.8 ± 0.2. The exptl. system showed promise as a ref. to compare metal ion speciation methodol.

NaSCN and an alkanolamine or alkanolamino acid. .
