I (2 ′ 10 mg/kg, i.v.) produced a rapid and long-lasting redn. (-30%) of striatal dopamine [51-61-6], whereas the dopamine levels were only transiently reduced in mesencephalon and frontal cortex. Homovanillic acid [306-08-1] and DOPAC [102-32-9] were initially reduced (-50 to -70%) in striatum; whereas a marked recovery was obsd. in the chronic stage. I also induced a long-term redn. of noradrenaline [51-41-2] in the striatum and frontal cortex whereas 5-HT [50-67-9] and 5-HIAA [54-16-0] levels were essentially unaltered.In this experiment, several chemicals are used like 54-16-0 and 51-61-6 The data indicate a neurotoxic action of I on both dopamine and noradrenaline nerve terminals in mouse brain. The results are discussed in light of the potential use of I as a denervation tool in Parkinsonism model. .

Curvilinear Scatchard plots were obsd. for both melanins, indicating that >1 binding class must be implicated. The data for the binding of MPTP to beef eye melanin could be fitted by the assumption of 2 classes of binding sites and to dopamine melanin by 3 classes of binding sites. Beef eye and dopamine melanin contained a small no. of binding sites with a high assocn. const. and a great no. of binding sites with a low assocn. const. The concordance between the assocn. consts. indicates a binding to identical sites on the 2 melanins. The total binding capacity of beef eye melanin (0.53 mmol/mg melanin) was ~2 times that of dopamine melanin (0.27 mmol/mg melanin).
