DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) To establish a bioassay for environmental toxicants, optimal conditions for the redn. of 2,3,5-triphenyltetrazolium chloride (TTC) [298-96-4] to triphenylformazan (TPF) [20150-01-0] by dehydrogenase [9035-82-9] of T. pyriformis (protozoa) were investigated. Optimal TTC concn. was 0.There are some reagents with their cas registry numbers 9035-82-9 and 20150-01-0 are used in this study.1% and optimal incubation was at 41° and pH 7.85 for 45 min. The TTC redn. activity of Tetrahymena cells increased with incubation time. For a given incubation time, a plot of cell no. vs. TPF absorbance was linear. The variations of TPF absorbance was 9.7 and 11.5% at 48 and 72 h after incubation, resp. Similar results were obtained for the cell count and protein concn. The cell no. vs. TPF absorbance, protein content vs. TPF absorbance and cell no. vs. protein concn. were highly correlated. .

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Four short-term microbiol. toxicity screening tests were compared using the following test chems.: 3,5-dichlorophenol [591-35-5], cetyltrimethylammonium chloride [112-02-7], Na lauryl sulfate [151-21-3], phenol [108-95-2], CuSO4, HgCl2, and ZnSO4. These 7 chems. represent a wide range of toxicity. The methods examd. were Beckman's Microtox system, the Spirillum volutans motility test, inhibition of respiratory activity of activated sludge, and inhibition of activated sludge triphenyltetrazolium chloride [298-96-4]-dehydrogenase [9035-82-9] activity. Each method has its own toxicity sensitivity pattern, and among the substances tested, only HgCl2 and phenol were ranked equally by the 4 methods as the most and the least toxic chem., resp. In a tentative ranking of the methods, according to sensitivity, the Microtox test came out as the most sensitive test, followed by the Spirillum test, which in turn appeared more sensitive than the 2 sludge tests. This ranking has meaning only in a statistical sense, however, and the variable nature of the results support the philosophy that for assessing toxicity more thoroughly, a battery of several tests is required. .
