DOCUMENT TYPE: Conference CA Section: 1 (Pharmacodynamics) In dogs with exptl. renal hypertension, the resistance of highly toned blood vessels of the hindlimb was decreased by the myotropics, papaverine [58-74-2] (1 mg/kg), No-Spa [14009-24-6] (1 mg/kg), and euphyllin [317-34-0] (3 mg/kg), to a lesser extent than that of the low-toned blood vessels of the brain. The neurotropics, reserpine [50-55-5], benzohexonium [971-60-8], and methylanogalanthamine [18126-83-5], decreased perfusion pressure in hindimb to a greater extent than in brain vessels. Caroted arch denervation increased the effect of myotropics on hindlimb but not brain vessels.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Patients with cirrhosis given single i.v. doses of theophylline ethylenediamine (I ethylenediamine) [317-34-0] had a prolonged plasma half-life (mean, 25.6 h vs. 6.7 h in controls) and a decreased plasma clearance (mean, 0.042 L/kg/h vs. 0.062 L/kg/h). Vols. of distribution of I in the cirrhotic patients did not substantially differ from normal. Plasma I binding in patients with cirrhosis averaged 36.8% as compared with 52.6% in normal subjects. There was no correlation between any lab. test of liver function and the plasma I half-life, except for serum albumin. The variable capacity to eliminate I precludes the use of usual maintenance dose schedules for bronchodilation in cirrhosis.
