(HPLC) assay was developed for the quantitation of 2 structurally similar and highly active anticancer drugs, etoposide (I) [33419-42-0] and teniposide (II) [29767-20-2], and their potential metabolites. The assay utilizes electrochem. detection, which imparts specificity and sensitivity sufficient to detect 320 ng/mL in plasma, urine, and cerebrospinal fluid (CSF). The mean assay coeffs.Several substances with their cas registry numbers 29767-20-2 and 89269-95-4 may be metioned in this study. of variation were 5.1 and 8.1% for teniposide (10 mg/mL) and etoposide (5 mg/mL), resp. The extn. efficiencies were 86% for etoposide, 70% for its hydroxy acid metabolite [89269-95-4], 66% for teniposide, and 54% for the hydroxy acid of teniposide [89269-94-3]. The correlation coeff. of the multilevel std. curve was 30.995 over the concn. range of 0.05-50 mg/mL for the parent and metabolites extd. from plasma. This method was used to det. the concns. of the parent drugs and their metabolites in the plasma, urine, and CSF of patients with cancer. .

The effect of disulfiram, an aldehyde dehydrogenase [9028-86-8] inhibitor, and its metabolite, diethyldithiocarbamate, on I-induced DNA damage in mouse leukemia L-1210 cells was assayed by using the alk. elution technique. Disulfiram and diethyldithiocarbamate inhibited I-induced single-strand breakes in DNA. Both compds. also reduced I-induced cytotoxicity. Oxalic acid pyrophosphate and malonic acid, competitive inhibitors of succinate dehydrogenase [9002-02-2], and the naturally occurring dehydrogenase substrates, succinic acid, b-glycerophosphate, and isocitric acid, also blocked the effects of I. Free-radical scavengers were also studied. Na benzoate was particularly effective in preventing drug-induced DNA damage and cytotoxicity, but a no. of other scavengers were not. Results were consistent with the hypothesis that I is activated by an enzyme such as a dehydrogenase which transforms it into an active intermediate resulting in DNA damage and, consequently, cell death. .
