The hybrid gene created by this fusion encodes a polypeptide which has 25 addnl. amino acids on the N terminus of the HSV-1 TK protein and phenotypically complements a tdk- mutation of E. coli. This fusion polypeptide has been characterized by maxicell, immunopptn., and native gel techniques, and its activity is inhibited by anti-HSV-1 antibody. In a tk expressor strain contg. a F' lacIq (which overproduces the lactose repressor), the isopropyl-b-D-thiogalactoside [367-93-1] causes >1000-fold coordinate induction of the plasmid-encoded TK and chromosomal b-galactosidase activities. Pulse-chase induction demonstrates the fused TK polypeptide to be as stable as b-galactosidase. HSV-1 tk-specific RNA isolated from this bacterial strain has a short half-life characteristic of bacterial messages.

In the presence of isopropyl-b-D-thiogalactoside [367-93-1] the system was derepressed (repressor inactivated) to enhanced prodn. of penicillinase (6000 units/mL culture medium compared with 60 units/mL in the absence of IPTG) from the hybrid promoter. Similarly, B. subtilis served as a host for the controlled (IPTG-induced) expression of a cloned human leukocyte interferon A from a spac-1 promoter/lac operator on a plasmid with a downstream lacI repressor-encoding sequence. The spac-1 promoter is similar to the B. subtilis pac-1 promoter except that the RNA polymerase recognition site is derived from a B. subtilis phage promoter.
