DOCUMENT TYPE: Journal CA Section: 9 (Biochemical Methods) Section cross-reference(s): 34 15N-labeled amino acids were synthesized from the respective .alpha.-keto acids in a reaction mixt. contg. borate buffer, pH 9.0, glucose-6-phosphate, either Na .alpha.-ketobutyrate or Na .alpha.-ketomethiobutyrate or Na .alpha.-ketoiso valerate, NADP, 15NH4Cl (95 atom% 15N), glucose-6-phosphate dehydrogenase, and glutamate dehydrogenase. Reactions were run for 36 h at 30.degree. in a sealed vessel, with the pH being kept at 8.5-9.0 by the addn. of KOH. The reaction mixt. then was boiled, filtered, and purifd. on an Amberlite IR 120 H+ cation-exchange column. The amino acid eluate was concd. and crystallized with EtOH-Et2O. The yields were 68, 41, and 42% of theory for L-.alpha.-aminobutyric acid, L-methionine, and L-valine, resp.; the acids were >99% pure by gas chromatog., and contained 95 atom% 15N. .
