An attempt was made to prevent this biochem. injury by a daily administration of an anabolic steroid: 19-nortestosterone phenylpropionate (I) [62-90-8] (25 mg/kg/24 h, i.p.), to female rates, 8 days before the administration of 2 steatogenic agents: white phosphorus (WP) (10 mg/kg, s.c.) and Amanita phalloides ext. (AP) (0.5 mL/kg, i.p.). The animals were killed 36 h (WP) or 18 h (AP) after poisoning and hepatic lipid components were detd. In both cases, the TG accumulated were responsible for the fatty liver, with 5-6 fold increase as compared with controls. The administration of I provoked a decrease of liver TG in both intoxications, but with different intensity: very marked with AP, moderately marked with WP. Apparently, a much more specific protective effect of I in AP fatty liver, related to the stimulation of RNA polymerase II, inhibited by a-Amanitin; in WP fatty liver, the moderate effect of I may be due to a nonspecific stimulation of the biosynthesis of liver apolipoproteins.

The antiandrogenic effect of I depended on its dose as well as on the dose of the agonist. A modification of the method used to assess biol. activity of antiandrogens consisted of a single administration of anabolic prepns. with prolonged activity and 7-day administration of the antiandrogen to castrated animals.
