DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) In model expts. on the protective mechanism of silymarin [65666-07-1] against hepatotoxic peroxidn. processes and free radicals (e.g., as from CCl4), 5-20 mM silymarin inhibited O uptake by isolated rat liver mitochondria and microsomes in lipid-peroxidative systems (Fe2+-ascorbate and NaDPH-Fe3+-ADP, resp.). Two water-sol. derivs. of 1 of the components of silymarin, silybin N-methylglucamine [51148-13-1] and silybin dihemisuccinate [67113-62-6], in decreased order of activity, were less effective than silymarin in inhibiting peroxidn. in the mitochondrial and microsomal systems, indicating the importance of the hydrophobic properties of silymarin for its antiperoxidative effect. Silymarin reacted directly in vitro with the stable free radical 2,2-diphenyl-1-picrylhydrazyl [1898-66-4], as shown by spectrophotometry and ESR spectrometry. The stoichiometry of silymarin to free radical was 1.5-2:1. Silymarin can be included in the class of free-radical scavengers.
