DOCUMENT TYPE: Journal CA Section: 17 (Food and Feed Chemistry) Quant. anal. of each of the S-alk(en)yl-L-cysteine sulfoxides of onions was achieved by the following procedure. A MeOH-CHCl3-H2O ext. of onion tissue was purified by electrophoresis. Alk(en)yl-L-cysteine sulfoxides were sepd. by TLC on com. silica-gel plates and were reacted with ninhydrin, the spot intensity being measured by an integrating densitometer. The synthetic analog (±)-S-1-butyl-L-cysteine sulfoxide was used as an internal std. in each ext. In white bulb onion (cv. Southport White Globe), (±)-S-1-propyl-L-cysteine sulfoxide [17795-24-3] is the predominant flavor precursor at a concn. of 2.9 mg/g fresh wt. (±)-S-1-Methyl-L-cysteine sulfoxide [6853-87-8] and (±)-trans-S-1-propenyl-L-cysteine sulfoxide [16718-23-3] are present at the lower concns. of 0.9 and 0.6 mg/g fresh wt., resp. Stable Bu, trifluoroacetyl derivs. of stds. and of the alk(en)yl-L-cysteine sulfoxides in exts., were formed. Subsequent gas chromatog.-mass spectrometry enabled parent ions and fragmentation patterns to be recorded and allowed the conclusive identification of the endogenous alk(en)yl-L-cysteine sulfoxides. .

Following the onset of kale feeding, all animals developed subclin. hemolytic anemia which stabilized by wk 6, and was more marked during wk 1-6 than during wk 7-12. Animals grazing low SMCO kale showed a less severe anemia than those grazing normal SMCO kale, which was assocd. with lower blood concns. of Me2S and Heinz bodies, and higher reduced glutathione and packed cell vol. levels. Live-wt. gain and wool growth were greater for lambs grazing low than normal SMCO kale during wk 1-6, corresponding to the period of most severe hemolytic anemia, but during wk 7-12 there were no differences between the 2 groups. SMCO content thus depressed kale feeding value, with most of the depression occurring in the 1st 6 wk of grazing; the lambs adapted to kale contg. 0.6% SMCO after 6 wk of feeding. Endocrine factors involved in this adaptation are discussed.
