DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The participation of a local anesthetic action in the spinal reflex inhibition produced by mephenesin-type muscle relaxants was examd. by comparing the local anesthetic effects (in vitro), the depressant effects on muscle afferent discharges (in situ), and the depressant effects on spinal reflexes (in situ) of the drugs in rats. At doses producing depression of spinal reflexes, mephenesin [59-47-2], tolperisone [728-88-1], and lidocaine [137-58-6] reduced the frequency of afferent discharges from the muscle. The order of reducing afferent discharges of these drugs corresponded to that of their conduction-blocking activities in the isolated sciatic nerve of rats. Apparently, a membrane-stabilizing action participates in the spinal reflex inhibition produced by mephenesin-type muscle relaxants. Baclofen [1134-47-0] (nonmephenesin type) did not show any local anesthetic action.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Baclofen [1134-47-0] and dantrolene [7261-97-4] intraduodenal (i.d.) administration suppressed sustained rigidity of forelimbs in rats with cerebral ischemia. ED50 of baclofen was 2.9 mg/kg and of dantrolene was 22 mg/kg. Baclofen, dantrolene, and diazepam [439-14-5] reduced the phase rigidity of the decerebrated animals with i.d. ED50 of 6.2, 140, and 1.4 mg/kg, resp. Both rigidities were insensitive to chlorodiazepoxide [58-25-3] and tolperisone [728-88-1]. Except for tolperisone, these drugs also produced muscle relaxation in intact rats and mice. The ED50 values of orally administered baclofen, dantrolene, diazepam, chlordiazepoxide, and tolperisone were 5.6, 15.0, 2.1, and 44.0 mg/kg, resp. in mice. .
