DOCUMENT TYPE: Journal CA Section: 18 (Animal Nutrition) As a measure of lipid peroxidn., pentane expired in the breath of rats loaded with an accumulated dose of either 100 mg or 186-200 mg of Fe injected i.p. as Fe dextran [9004-66-4] was measured over a 7-8 wk period, and the effect on pentane prodn. of feeding antioxidant-supplemented diets was detd. By the 7th week of feeding the diets, rats fed 0.3% L-ascorbic acid [50-81-7] produced 17% less pentane than did rats fed the basal antioxidant-deficient diet, whereas rats fed 0.004% dl-a-tocopherol acetate [52225-20-4] produced 92% less. After being fed the basal diet for 7 wk, Fe-loaded rats produced 76 pmol pentane/100 g body wt/min. When synthetic antioxidants were added to the diet at a concn. of 0.25%, the order of effectiveness in decreasing pentane prodn. after 1 wk was: N,N'-diphenyl-p-phenylenediamine [74-31-7] > ethoxyquin [91-53-2] > BHA [25013-16-5] > BHT [128-37-0] > propyl gallate [121-79-9] u no antioxidant. After removal of either ethoxyquin or N,N'-diphenyl-p-phenylenediamine from the diets for 1 wk, pentane prodn. increased to a high level. The total amt. of lipid-sol. 128-37-0 and 121-79-9 which are cas registry numbers of substances are two of reagents here. fluorophores in individual spleens of rats fed N,N'-diphenyl-p-phenylenediamine, ethoxyquin, dl-a-tocopherol acetate, ascorbic acid, and no antioxidant were correlated significantly with the corresponding total integrated amt. of pentane produced by the individual rats over the 7-8 wk period. This study has provided some of the most direct evidence to date that lipid peroxidn. is assocd. with Fe overload in vivo. .
