DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Benzonal (I) [744-80-9], benzobamil (II) [7279-57-4], and phenobarbital (III) [50-06-6] were given to rats (orally, 1/20 LD50 daily) for 4, 14, or 30 days. Short-term administration of I and II induced liver synthesis of mixed-function oxygenases, liver hypertrophy, proliferation of hepatocyte endoplasmic reticulum, and increases in microsomal cytochrome P 450 [9035-51-2] and cytochrome b5 [9035-39-6]. It also increased the rate of hydroxylation of hexobarbital and aminopyrine, without affecting the rate of spontaneous peroxidn. of lipids. Month-long application of I, II, and III, however, led to normalization of liver mass, microsomal protein, and (in the case of I and III) cytochrome P 450. Cytochrome b5 levels remained elevated. Month-long administration of I and III led to decreased oxidn. of aminopyrine and hexobarbital, but administration of II activated the metab. of these xenobiotics and increased cytochrome P 450. Lipid peroxidn. was below normal at this time. I is recommended as a microsomal inducer in treatment of disease such as hyperbilirubinemia. II, combining a weak inductive effect with marked hepatoprotectant effect, is recommended for treatment of epileptics with impaired liver function.

The liver enzyme inducers prevented the development of pathol. changes in the adrenal glands due to I. The possible use of the inducers to diminish I toxicity without altering its antibacterial activity is discussed.
