- Hydroamination of ethylene by aniline: Catalysis in water

The platinum-catalyzed and halide-promoted hydroamination of ethylene with aniline is reported for the first time in the presence of simple sodium halides in water. Compounds K2PtX4 (X = Cl or Br), PtX2 or PtX4 (0.3% mol) in the presence of an aqueous solution of excess NaX and aniline under ethylene pressure (25 bar) affords N-ethylaniline with 60-85 turnovers after 10 h at 150°C. The best result (TON = 85) was obtained in the presence of excess NaBr, whereas a slightly lower activity was observed with NaCl (60 cycles) and practically no activity with NaF or NaI (2-4 cycles). The reaction also produces N,N-diethylaniline (up to 1 cycle) and 2-methylquinoline (up to 8 cycles) as by-products. The influence of added H + and different oxidizing agents was also examined.

One-pot tandem synthesis was developed for substituted quinolines (in up to 97% yields) involving a selective catalytic oxidation of primary amines to aldehydes and their condensation with arylamines under the action of a dispersion of Fe(CrO2)2 and water solution of H2O2 at room temperature. The stage of catalytic oxidation of alcohols was accelerated by photoactivation. A presumable mechanism of the photoactivated tandem synthesis of 2-methylquinoline was suggested. Catalyst Fe(CrO2)2 was prepared by photochemical synthesis.

A simple and general protocol was developed for selective conversion of amides into amines. Amides were converted into amide acetals and imido esters by O-alkylation and then hydrogenated without isolation into amines under very mild reaction conditions over standard hydrogenation catalysts. Triethyloxonium tertafluoroborate, methyl trifluoromethanesulfonate, dimethyl sulfate and ethyl chloroformate were validated as alkylating agent. The synthetic utility of this approach was demonstrated by the selective carbonyl reduction of peptide groups. Carbonyl reduction of peptide group proceeds chemoselective without racemization of the neighboring chiral center. (Figure presented.).

The selectivity of the catalytic reaction between aniline and ethylene in the presence of the Brunet catalyst (PtBr2/Br-) shifts from the hydroamination product N-ethylaniline to the heterocyclization product 2-methylquinoline (quinaldine) when conducted in the presence of PPh3 (1 equiv per Pt atom). Condition optimization revealed that this process works best in the absence of any halide salt additive, that it is essentially insensitive to the nature of the halide in PtX2, that the best promoter in the PMexPh3-x series is PPh3 when used in strictly stoichiometric amounts, and that the 4-RC6H 5NH2 (R = nBu, Cl, OMe, NMe2) substrates are equally converted albeit less efficiently. Slight dilution of the system with THF or toluene slightly improves the activity, and a kinetic profile shows the presence of an induction phase and a deactivating step, which however does not involve reduction to metallic platinum contrary to the PPh3-free Brunet catalyst. Mechanistic considerations are presented.

A DFT/B3LYP study with inclusion of solvent and temperature effects has probed the olefin activation mechanism for the intermolecular hydroamination of ethylene and 1-hexene by aniline derivatives catalyzed by the PtX 2/X- system on the basis of a variety of experimental results, including new experiments on catalyst deactivation. For ethylene and aniline, the calculated ΔG?cycle between the resting state [PtX3(C2H4)]-, 1X, and the TOF-determining transition state of the C-H reductive elimination from [PtX3(H)-(CH2CH2NHPh)]-, TS2 X, is slightly smaller for X = Br than for Cl or I. The ΔG?cycle decreases as the aniline basicity decreases. For the slightly less efficient hydroamination of 1-hexene, ΔG? cycle is greater than that for the hydroamination of ethylene, with a preference for the Markovnikov addition, in agreement with experiment and with essentially equivalent ΔG?cycle values for the Br and I systems. In general, the results of the calculations are in agreement with the experimental observation. A clear-cut comparison of trends is hampered by the small energy differences and by the possibility, proven in certain cases, that the reaction parameters under investigation affect the catalyst degradation rate in addition to its intrinsic activity. Extrapolation of the computational study to the fluoride system suggests that this should be even more active. However, experimental studies show that this is not the case. The reason for this anomaly has been traced to the basicity of the fluoride ion, which triggers more rapid catalyst decomposition. A bonding analysis of 1X indicates a significant push-pull π interaction between the C2H4 and the trans-F ligand.

Nitroarenes are reductively converted into quinoline derivatives with aliphatic alcohols in the presence of a catalytic amount of ruthenium compound at 180 degree C. Ruthenium(III) chloride is the most effective catalyst. The reaction of nitrobenzene with 1-propanol and 1-butanol gave 2-ethyl-3-methylquinoline and 3-ethyl-2-propylquinoline in 65 and 70% yields respectively. p-Methoxynitrobenzene gave 3-ethyl-6-methoxy-2-propyquinoline in 70% yield with 1-butanol. The reaction appears to include the redox reaction between the nitroarenes and the alcohols, that is, a catalytic hydrogen transfer reaction which generates the aminoarenes and aldehydes. Thus, the alcohol plays roles as both a reductant and an aldehyde precursor.

The catalytic system "PtBr2(0.3 mol%)/2P(OR) 3/10nBu4PBr" (R = alkyl) discovered recently in our group, allows good to excellent catalytic activities for the intermolecular hydroamination of ethylene and higher α-olefins (1-hexene) with aniline type amines to give the expected N-ethyl- (1) and N,N-diethyl-anilines (2) along with 2-methyl-quinoline (3). A poisoning effect of alkyl and aromatic phosphines has been observed. This effect could be minimised using small amounts of molecular iodide which reacts with phosphorus ligands to form non-coordinating well-described ionic species. Interestingly, beneficial effects of added P(OR)3 (R = alkyl) have been pointed out and further investigated. In this way, a new potential family of Arbuzov-type phosphorus-metal complexes has been suggested to be responsible for good catalytic activities found when using P(OR)3 alkyl phosphites rather than PR3 (R = alkyl, aromatic) and P(OPh)3 co-catalysts.

Metal nanoparticles (NPs) encapsulated in hollow nanostructures hold great promise for a variety of applications. Herein, we demonstrate a new concept where covalent organic frameworks (COFs) doped with metal cations can be readily used as novel precursors for the in situ encapsulation of metal NPs into N doped hollow carbon spheres (NHCS) through a controlled carbonization process. The obtained Pd@NHCS composites show a significantly enhanced catalytic activity and selectivity in the hydrogenation of nitrobenzene in ethanol and oxidation of cinnamyl alcohol compared with that of the conventional Pd/N-C and commercial Pd/C catalysts. The excellent catalytic performance should be related to the synergism of the porous hollow spheric structure, highly dispersed Pd NPs, and uniform distribution of N dopants on the materials. We believe that this newly developed methodology could be extended to the synthesis of other metal NPs@NHCS composites for a variety of advanced applications.

- Vapor-phase synthesis of 3-methylindole from glycerol and aniline over zeolites-supported Cu-based catalysts

Zeolites-supported Cu-based catalysts for the vapor-phase synthesis of 3-methylindole from glycerol and aniline were investigated. The catalysts were characterized by N2 adsorption, XRD, NH3-TPD and TG-DTA techniques. The results indicated that Cu/NaY catalyst exhibited higher activity and selectivity than Cu/5A, Cu/HZSM-5 or Cu/MCM-41 due to better dispersion of copper particles and larger amount of weak acid sites on the catalyst. The addition of Na2O to Cu/NaY not only improved the selectivity of the catalyst efficiently, but also inhibited the deactivation of the catalyst. Over Cu/NaY-Na2O, the selectivity of 3-methylindole reached 72.3% and the yield of the aim product was up to 50.4%. The characterizations revealed that Na2O promoter could decrease the amount of middle-strong acid sites of the catalyst remarkably as well as prevent from the formation of the coke. Furthermore, a probable catalytic mechanism for the synthesis of 3-methylindole from glycerol and aniline was proposed.

These triazenes are highly stable, and even in strongly acidic medium (0.5 molL -1 H2SO4) they are only slowly decomposed back to the diazonium ion and substituted anilinium ion (for the N-ethyl derivative in 0.5 mol·L-1 H2SO4 at 25 °C, t1/2 ≈ 7 h). On either direct irradiation in water or triplet-sensitized irradiation in acetone, pyrimidines 1a-c undergo hydrogen abstraction from an nπ* triplet state, fragmentation, and rearomatization to form 5 (eq 1). Similarly, direct irradiation of quinolines 2a-c in benzene or tert-butyl alcohol furnishes 6 from an nπ* singlet state. Quantum yields for products (Φ5 and Φ6) and Stern-Volmer quenching studies on these reactions (Tables I and II) provide mechanistic information concerning abstraction by aromatic nitrogen. Unlike Φ for abstraction by carbonyl triplets, Φ5 and Φ6 do not increase in hydrogen-bonding solvent, and there is little rate difference in transferring primary, secondary, and tertiary hydrogen in the initial step. Singlet and triplet abstractions show little difference in selectivity, but the singlet reaction is ? 104 faster. Concentration studies show that fragmentation of 1a-c is self-quenched, while reaction of 2a-c is enhanced with increasing concentration.

A green route for the vapor-phase synthesis of quinoline from glycerol and aniline was developed in this work, employing Ni/mesoporous beta zeolite (denoted as Ni/Hβ-At) as a catalyst. The mesoporous beta zeolite was prepared by alkaline treatment. Various influencing factors were systematically investigated. Both mesopores and the type of acid sites of the catalyst played important functions in catalytic activity for the synthesis of quinoline. Mesopores facilitated the transport of bulky products from internal surface of the catalyst. Meanwhile, weak Br?nsted acid sites favored the dehydration of glycerol to acrolein and the existence of Lewis acid sites could accelerate the formation of quinoline. The Ni/Hβ-At catalyst exhibited the highest catalytic activity; and as high as a 71.4% yield of quinoline was obtained under the optimized reaction conditions. An enhanced ability of anti-deactivation was also displayed, due to the existence of mesopores on the Ni/Hβ-At catalyst facilitating the transport of bulky products and restraining the deposition of the coke. Meanwhile, it was found that the coke was main reason leading to catalyst deactivation and its performance was basically regenerated. The catalytic properties were slightly lower after 3 reaction-regeneration cycles. Finally, a feasible reaction pathway was proposed on the basis of the various products.

These complexes can be easily filtered out from the reaction medium and reused up to five times without significant loss of catalytic activity.

Complex 1 could be transformed to 2 when reacted with NaOtBu or NEt3 via -OH deprotonation. These six complexes were tested as catalysts for mono-N-alkylation of amines with trialkyl amines, and complex 1 exhibited highest activity. - Oxygen-implanted MoS2 nanosheets promoting quinoline synthesis from nitroarenes and aliphatic alcohols via an integrated oxidation transfer hydrogenation-cyclization mechanism

We herein report that MoS2 with oxygen-implanting modification (O-MoS2) can work as a multifunctional catalyst to achieve the one-pot quinoline synthesis from basic nitroarenes and aliphatic alcohols. Different from common knowledge that the application of MoS2-based catalysts and above quinoline synthesis need anaerobic conditions, we conduct the heterogeneous catalysis under an unusual air atmosphere. Catalyst characterization and experimental results indicate that the MoOx clusters implanted in the MoS2 skeleton, not the coordinatively unsaturated Mo sites (CUS Mo), dominate the generation of quinolines. By overturning the catalysis perception that O2 adsorption on MoSx can deactivate the MoS2-based catalysts using an efficient method for in situ healing of the MoOx structure in O-MoS2 and protecting the O-MoS2 catalyst by inhibiting unwanted MoOx elimination with extra H*, we innovatively introduce O2 into the quinoline synthesis. The robust O-MoS2 can be consecutively used ten times without regeneration and it offers 69-75% yields of 2-methylquinoline from nitrobenzene and ethanol. Furthermore, different from the traditional transfer hydrogenation-condensation mechanism, an integrated oxidation-transfer hydrogenation-cyclization mechanism is proposed over the O-MoS2 catalyst.

- Highly Active Ni Nanoparticles on N-doped Mesoporous Carbon with Tunable Selectivity for the One-Pot Transfer Hydroalkylation of Nitroarenes with EtOH in the Absence of H2
-
Cost-effective and environmentally friendly conversion of nitroarenes into value-added products is desirable but still challenging. In this work, highly dispersed Ni nanoparticles (NPs) supported on N-doped mesoporous carbon (Ni/NC-x) were synthesized via novel ion exchange-pyrolysis strategy. Their catalytic performance was investigated for one-pot transfer hydroalkylation of nitrobenzene (NB) with EtOH in absence of H2. Interestingly, the catalytic performance could be easily manipulated by tuning the morphology and electronic state of Ni NPs via varying the pyrolysis temperature. It was found that the Ni/NC-650 achieved 100 % nitrobenzene conversion and approx. 90 % selectivity of N,N-diethyl aniline at 240 °C for 5 h, more active than those of homogeneous catalysts or supported Ni catalysts prepared by impregnation (Ni/NC-650-IM, Ni/SiO2). This can be ascribed to the higher dispersion and better reducibility as well as richer surface basicity of the catalyst. More interestingly, the Ni/NC-650 catalyst achieved complete conversion of various nitroarenes, yielding imines, secondary amines, or tertiary amines selectively by simply controlling the reaction temperature at 180, 200 and 240 °C, respectively. The one-pot hydrogen-free process with non-noble metal catalysts, as demonstrated in this work, shows great promise for selective conversion of nitroarenes with ethanol to various anilines at industrial scale, from an economic, environmental, and safety viewpoint.

- A highly efficient Co-based catalyst fabricated by coordination-assisted impregnation strategy towards tandem catalytic functionalization of nitroarenes with various alcohols
-
A well-defined hexamethylenetetramine (abbreviated as HMTA) based two-dimensional (2D) MOFs metalloligand (termed Zn-HMTA), with free uncoordinated tertiary amine groups, has been synthesized via solution diffusion method for the first time. The crystal structure of 2D Zn-HMTA metalloligand was determined by the single crystal X-ray diffraction (SCXRD). The SCXRD and X-ray photoelectron spectroscopy (XPS) analyses have revealed that the 2D Zn-HMTA metalloligand is rich in- free tertiary amine groups, which are of strong coordination ability to transition metal ions (e.g. Ni2+, Co2+, Zn2+, Cu2+). As a result, a 2D bimetallic Co@Zn-HMTA MOFs was synthesized via coordination-assisted impregnation (CAI) strategy attributed to the unique feature of strong coordinated ability of free tertiary amine groups. Furthermore, a series of self-supported Co-ZnO-CN nanocatalysts were afforded upon the as-synthesized Co@Zn-HMTA MOFs served as a self-sacrificial template for pyrolysis at different temperatures. The optimized catalyst (termed as Co-ZnO@CN-CAI) demonstrated the excellent catalytic performance for hydrogenation-alkylation tandem reaction in comparison with the classic ZnO@CN composite (derived from Zn-HMTA MOFs) supported metallic Co catalyst (Co-ZnO@CN-IWI) prepared by incipient wetness impregnation method. Moreover, the kinetic study was also performed to confirm that the alkylation is the rate-determining step in the hydrogenation-alkylation tandem reaction. The origin of enhanced catalytic performance of Co-ZnO@CN-CAI and the role of Co@Zn-HMTA MOFs precursor have been explored by way of various characterizations, e.g. HADDF-STEM-EDS, SEM-EDS, 13C MAS NMR, XRD, Raman and XPS, etc. It is anticipated that the prepared low-cost and easily prepared 2D Zn-HMTA metalloligand will become a general template for synthesis of highly self-supported catalysts with coordination-assisted impregnation strategy (CAI) for various catalytic reactions.

- Nanosized CdS as a Reusable Photocatalyst: The Study of Different Reaction Pathways between Tertiary Amines and Aryl Sulfonyl Chlorides through Visible-Light-Induced N-Dealkylation and C-H Activation Processes

It has been found that the final products of the reaction of sulfonyl chlorides and tertiary amines in the presence of cadmium sulfide nanoparticles under visible light irradiation are highly dependent on the applied reaction conditions. Interestingly, with the change of a reaction condition, different pathways were conducted (visible-light-induced N-dealkylation or sp3 and sp2 C-H activation) that lead to different products such as secondary amines and various sulfonyl compounds. Remarkably, all of these reactions were performed under visible light irradiation and an air atmosphere without any additive or oxidant in benign solvents or under solvent-free conditions. During this study, the CdS nanoparticles as affordable, heterogeneous, and recyclable photocatalysts were designed, successfully synthesized, and fully characterized and applied for these protocols. During these studies, intermediates resulting from the oxidation of tertiary amines are trapped during the photoinduced electron transfer (PET) process. The reaction was carried out efficiently with a variety of substrates to give the corresponding products at relatively short times in good to excellent yields in parallel with the use of the visible light irradiation as a renewable energy source. Most of these processes are novel or are superior in terms of cost-effectiveness, safety, and simplicity to published reports.

- Catalyst-free photoinduced selective oxidative C(sp3)-C(sp3) bond cleavage in arylamines

Due to the directional nature of sp3-hybridized orbitals and the absence of π-orbitals, the oxidative cleavage of the kinetically and thermodynamically stable C(sp3)-C(sp3) bond is extremely difficult and remains scarcely explored. In this work, under the double argument of quantum mechanics (QM) computations and meticulous experiments on our well-designed C-C single bond cleavage mechanism, we discovered a means of photoinduced selective oxidative C(sp3)-C(sp3) bond cleavage in arylamines, easily achieved by simple visible light irradiation using O2as a benign oxidant under very mild conditions. The utility of our methodology was demonstrated by the C(sp3)-C(sp3) bond cleavage in morpholine/piperazine arylamines with excellent functional group tolerance. Importantly, our methodology is noteworthy, not only in that it does not require any catalysts, but also in that it provides valuable possibilities for the scalable functionalization of clinical drugs and natural products.

- Novel hybrid conjugates with dual estrogen receptor α degradation and histone deacetylase inhibitory activities for breast cancer therapy
-
Hormone therapy targeting estrogen receptors is widely used clinically for the treatment of breast cancer, such as tamoxifen, but most of them are partial agonists, which can cause serious side effects after long-term use. The use of selective estrogen receptor down-regulators (SERDs) may be an effective alternative to breast cancer therapy by directly degrading ERα protein to shut down ERα signaling. However, the solely clinically used SERD fulvestrant, is low orally bioavailable and requires intravenous injection, which severely limits its clinical application. On the other hand, double- or multi-target conjugates, which are able to synergize antitumor activity by different pathways, thus may enhance therapeutic effect in comparison with single targeted therapy. In this study, we designed and synthesized a series of novel dual-functional conjugates targeting both ERα degradation and histone deacetylase inhibiton by combining a privileged SERD skeleton 7-oxabicyclo[2.2.1]heptane sulfonamide (OBHSA) with a histone deacetylase inhibitor side chain. We found that substituents on both the sulfonamide nitrogen and phenyl group of OBHSA unit had significant effect on biological activities. Among them, conjugate 16i with N-methyl and naphthyl groups exhibited potent antiproliferative activity against MCF-7 cells, and excellent ERα degradation activity and HDACs inhibitory ability. A further molecular docking study indicated the interaction patterns of these conjugates with ERα, which may provide guidance to design novel SERDs or PROTAC-like SERDs for breast cancer therapy.

- A Highly Dispersed Copper Nanoparticles Catalyst with a Large Number of Weak Acid Centers for Efficiently Synthesizing the High Value-Added 3-Methylindole by Aniline and Biomass-Derived Glycerin

Abstract: An excellent catalyst with a large number of weak acid centers and highly dispersed copper nanoparticles embedded in mesoporous SBA-15 carrier was successfully constructed for the purpose of efficient conversion of aniline with biomass-derived glycerin to the high value-added 3-methylindole, in which the catalyst of Cu/SBA-15 was modified with Al2O3, La2O3 and CoO in sequence. The modified carrier and the copper-based catalysts were studied by scanning electron microscopy and energy-dispersive X-ray (SEM–EDX) spectroscopy, nitrogen physical adsorption, ammonia temperature programmed desorption (NH3-TPD), hydrogen temperature programmed reduction (H2-TPR), powder X-ray diffraction (XRD), transmission electron microscopy (TEM), thermogravimetric and differential thermal analysis (TG–DTA) and inductively coupled plasma (ICP) emission spectroscopy. The research found that the Cu/CoO/La2O3/Al2O3/SBA-15 catalyst exhibited a very good catalytic performance with 3-methylindole yield up to 73.3% and selectivity reaching 86.4%. Besides, only a 3.9% yield decreased after the catalyst was circulated seven times. The characterizations revealed that Al2O3 could enhance the polarity of the carrier, thereby the interaction between the active component and the composite carrier was strengthened and the dispersion of copper was increased significantly. Adding La2O3 to Cu/SBA-15-Al2O3 could weaken the acidity and inhibit the formation of carbon deposits. CoO promoter could increase the number of weak acid centers, which was conducive to a good dispersion of active component and the high selectivity of 3-methylindole. - Linear Hydroaminoalkylation Products from Alkyl-Substituted Alkenes

The regioselective conversion of alkyl-substituted alkenes into linear hydroaminoalkylation products represents a strongly desirable synthetic transformation. In particular, such conversions of N-methylamine derivatives are of great scientific interest, because they would give direct access to important amines with unbranched alkyl chains. Herein, we present a new one-pot procedure that includes an initial alkene hydroaminoalkylation with an α-silylated amine substrate and a subsequent protodesilylation reaction that delivers linear hydroaminoalkylation products with high selectivity from simple alkyl-substituted alkenes. For that purpose, new titanium catalysts have been developed, which are able to activate the α-C?H bond of more challenging α-silylated amine substrates. In addition, a direct relationship between the ligand structure of the new catalysts and the obtained regioselectivity is described.

- Electroactivated alkylation of amines with alcohols: Via both direct and indirect borrowing hydrogen mechanisms

A green, efficient N-alkylation of amines with simple alcohols has been achieved in aqueous solution via an electrochemical version of the so-called "borrowing hydrogen methodology". Catalyzed by Ru on activated carbon cloth (Ru/ACC), the reaction works well with methanol, and with primary and secondary alcohols. Alkylation can be accomplished by either of two different electrocatalytic processes: (1) in an undivided cell, alcohol (present in excess) is oxidized at the Ru/ACC anode; the aldehyde or ketone product condenses with the amine; and the resulting imine is reduced at an ACC cathode, combining with protons released by the oxidation. This process consumes stoichiometric quantities of current. (2) In a membrane-divided cell, the current-activated Ru/ACC cathode effects direct C-H activation of the alcohol; the resulting carbonyl species, either free or still surface-adsorbed, condenses with amine to form imine and is reduced as in (1). These alcohol activation processes can alkylate primary and secondary aliphatic amines, as well as ammonia itself at 25-70 °C and ambient pressure.

- Effective and selective direct aminoformylation of nitroarenes utilizing palladium nanoparticles assisted by fibrous-structured silica nanospheres

Abstract: Palladium nanoparticles (~ 1–3?nm, 0.4?wtpercent Pd) were uniformly distributed over the surface of fibrous silica nanospheres (KCC-1) modified via aminopropyltriethoxysilane using a fast and cost-effective palladium (II) chloride reduction process. The Pd nanoparticles (Pd NPs) distribution over the ensuing catalyst Pd/KCC-1-NH2 showed much more uniform distribution, and smaller size compared with the tedious hydrothermal reduction method. The morphological, chemical, and size analyses of Pd/KCC-1-NH2 by BET, UV–Vis spectra, XRD, HR-TEM, EDS and XPS analysis revealed that the succeeding material consist of a distinct fibrous silica nanospheres support adorn with Pd NPs. The resultant nanocatalyst was tested for the one-step reductive aminoformylation of aromatic nitro compounds using formic acid. A wide range of substituted nitroarenes including electron withdrawing, releasing, sterically hindered and multifunctional groups have been converted to corresponding aryl formamide in quantitative yields (yields up to 98percent) at moderate temperature (70?°C). Optimization study has proved that the 6 equivalent of formic acid is required and toluene was found to be the better solvent. The established practice is beneficial due to the use of formic acid as H2 source and formylating agent, easiness in handling of the catalyst and simple workup procedure with efficient catalyst reusability. - N -Arylation of (hetero)arylamines using aryl sulfamates and carbamates via C-O bond activation enabled by a reusable and durable nickel(0) catalyst

An effective and general aryl amination protocol has been developed using a magnetically recoverable Ni(0) based nanocatalyst. This new stable catalyst was prepared on Fe3O4@SiO2 modified by EDTA and investigated by FT-IR, EDX, TEM, XRD, DLS, FE-SEM, XPS, NMR, TGA, VSM, ICP and elemental analysis techniques. The reaction proceeded via carbon-oxygen bond cleavage of (hetero)aryl carbamates and sulfamates under simple and mild conditions without the use of any external ligands. This method demonstrated functional group tolerance in the N-arylation of various nitrogen-containing compounds as well as aliphatic amines, anilines, pyrroles, pyrazoles, imidazoles, indoles, and indazoles with good to excellent yields. - Continuous-Flow Amide and Ester Reductions Using Neat Borane Dimethylsulfide Complex

Reductions of amides and esters are of critical importance in synthetic chemistry, and there are numerous protocols for executing these transformations employing traditional batch conditions. Notably, strategies based on flow chemistry, especially for amide reductions, are much less explored. Herein, a simple process was developed in which neat borane dimethylsulfide complex (BH3?DMS) was used to reduce various esters and amides under continuous-flow conditions. Taking advantage of the solvent-free nature of the commercially available borane reagent, high substrate concentrations were realized, allowing outstanding productivity and a significant reduction in E-factors. In addition, with carefully optimized short residence times, the corresponding alcohols and amines were obtained in high selectivity and high yields. The synthetic utility of the inexpensive and easily implemented flow protocol was further corroborated by multigram-scale syntheses of pharmaceutically relevant products. Owing to its beneficial features, including low solvent and reducing agent consumption, high selectivity, simplicity, and inherent scalability, the present process demonstrates fewer environmental concerns than most typical batch reductions using metal hydrides as reducing agents.

- Diethylsilane as a Powerful Reagent in Au Nanoparticle-Catalyzed Reductive Transformations

Diethylsilane (Et2SiH2), a simple and readily available dihydrosilane, that exhibits superior reactivity, as compared to monohydrosilanes, in a series of reductive transformations catalyzed by recyclable and reusable Au nanoparticles (1 mol-%) supported on TiO2. It reduces aldehydes or ketones almost instantaneously at ambient conditions. It can be used in a one pot rapid reductive amination procedure, in which premixing of aldehyde and amine is required prior to the addition of the reducing agent and the catalyst, even in a protic solvent. An unprecedented method for the synthesis of N-arylisoindolines is also shown in the reductive amination between o-phthalaldehyde and anilines. In this transformation, it is proposed that the intermediate N,2-diphenylisoindolin-1-imines are reduced stepwise to the isoindolines. Finally, Et2SiH2 readily reduces amides into amines in excellent yields and shorter reaction times relative to previously known analogous nano Au(0)-catalyzed protocols.

The organoactinide-catalyzed (Cp*2ThMe2) hydroborated reduction of a wide range of tertiary, secondary, and primary amides to the corresponding amines/amine-borane adductsviadeoxygenation of the amides is reported herein. The catalytic reactions proceed under mild conditions with low catalyst loading and pinacolborane (HBpin) concentration in a selective fashion. Cp*2ThMe2is capable of efficiently catalysing the gram-scale reaction without a drop in efficiency. The amine-borane adducts are successfully converted into free amine products in high conversions, which increases the usefulness of this catalytic system. A plausible mechanism is proposed based on detailed kinetics, stoichiometric, and deuterium labeling studies.

- Direct N-Alkylation/Fluoroalkylation of Amines Using Carboxylic Acids via Transition-Metal-Free Catalysis

A scalable protocol of direct N-mono/di-alkyl/fluoroalkylation of primary/secondary amines has been constructed with various carboxylic acids as coupling agents under the catalysis of a simple air-tolerant inorganic salt, K3PO4. Advantageous features include 100 examples, 10 drugs and drug-like amines, fluorinated complex tertiary amines, gram-scale synthesis and isotope-labelling amine, thus demonstrating the potential applicability in industry of this methodology. The involvement of relatively less reactive silicon-hydride compared with the traditional reactive metal-hydride or boron-hydride species required to reduce the amide intermediates presumably contributes to the remarkable functional group compatibility. - A simple method for the synthesis of sulfonic esters

An efficient and simple approach for the direct synthesis of aryl and heteroaryl sulfonic esters was developed using DMS and DES as alkoxysulfonylation reagents. The reaction is operationally simple and scalable. This protocol does not require solvent, expensive catalysts, base, ligand additives or other reagents. A wide range of sulfonic esters were synthesized in moderate to good chemical yields. This method has the advantage of low cost, facile and tolerated a wide range of substrates.

- A strategy of two-step tandem catalysis towards direct N-alkylation of nitroarenes with ethanol via facile fabricated novel Co-based catalysts derived from coordination polymers

Three novel N-doped carbon supported Co/Co3O4 catalysts, namely, Co@CN-hmta, Co@CN-larg and Co-Co3O4@CN-bipy, with sheet-, worm-, honeycomb-like morphologies respectively, have been fabricated by the pyrolysis of well-defined coordination polymers (CPs). Upon the as-prepared catalysts were applied for the reaction of N-alkylation of nitroarenes with ethanol, a direct two-step tandem reaction is realized, in which the Co@CN-hmta delivers 100% conversion/selectivity of N-ethylaniline/N,N-diethylaniline from the direct N-alkylation of nitroarenes with ethanol. The kinetic studies were conducted to confirm that the N-alkylation of aniline with ethanol is the rate-determining step in the two-step tandem reaction. The SEM/EDX, XRD, Raman, TEM, XPS, and CO2-TPD characterization results have revealed that sizes and dispersion of metallic Co, amount of structural defects and surface Lewis basicity towards three catalysts can be tuned by changing the structures of Co-based CPs designed by different organic linkers, which may also help to understand the preparation of industrial catalysts on a molecular level. The optimized Co@CN-hmta catalyst is easily recycled by using the external magnet for successive reuses without any loss in both activity and selectivity. To the best of our knowledge, this is the first carbon-nitrogen species supported Co/Co3O4 catalysts derived from the CPs, which could effectively catalyzed the N-alkylation of nitroarenes with ethanol to produce the secondary amines and/or tertiary amines. This low-cost, recyclable and easy scale-up N-doped carbon supported catalyst may be of potential application in various heterogeneous catalytic reactions.

Synthesis of a Cp Ru complex bearing an NH 2 -functionalized N -heterocyclic carbene (C-N H) was achieved by treatment of CpRuBr(isoprene) with an equimolar amount of a silver complex, which was generated from Ag 2 O and 1-(2-aminoethyl)-3-methylimidazolium bromide, in CH 3 CN at room temperature. The new CpRuBr(C-N H) complex showed a higher catalytic performance than the related CpRuCl(P-N H) and CpRuCl(N-N H) complexes. In the reaction of N -arylcarboxamides, the amine products were obtained in satisfactory yields under mild temperature conditions.

- N–Alkylation of Amines Catalyzed by a Ruthenium–Pincer Complex in the Presence of in situ Generated Sodium Alkoxide

We report the use of ruthenium–NNN-pincer complexes of the type (R2NNN)RuCl2(PPh3) (R = tBu, iPr, Cy and Ph) for the catalytic N-alkylation of primary amines under solvent-free conditions. For the first time, the base that is required to promote these reactions is generated in situ from the alcohol by the use of sodium. The resulting sodium alkoxide regenerates the alcohol substrate while acting as the water scavenger thus mitigating the need of an additional base. Among the catalysts screened, (tBu2NNN)RuCl2(PPh3) (0.02 mol-%) gives very high turnovers and good yields at 140 °C. The (tBu2NNN)RuCl2(PPh3) catalyzed N-alkylation tolerates a variety of amine and alcohol substrates. While excellent turnover (29000) was obtained for the (tBu2NNN)RuCl2(PPh3) (0.002 mol-%) catalyzed alkylation of aniline with cyclohexyl methanol, the turnovers obtained in the corresponding catalytic methylation of p-anisidine was also very high (12000). The (tBu2NNN)RuCl2(PPh3) catalyzed reactions have also been accomplished under open-vessel conditions resulting in a net dehydrogenative coupling reaction. This protocol has been used to transform benzene-1,2-diamines to benzimidazoles with high productivity (12000 turnovers). DFT studies indicate that while β-hydride elimination is rate-determining (RDTS: 24.31 kcal/mol) for the alcohol dehydrogenation segment which is endothermic, insertion of the imine is rate-determining (RDTS: 11.26 kcal/mol) for its hydrogenation that is exothermic.

A formal deoxygenative hydrogenation of amides to amines with RuCl2(NHC)(PNHP) (NHC = 1,3-dimethylimizadol-2-ylidene, PNHP = bis(2-diphenylphosphinoethyl)amine) is described. Various secondary amides, especially NH-lactams, are reduced with H2 (3.0-5.0 MPa) to amines at a temperature range of 120-150 °C with 1.0-2.0 mol % of PNHP-Ru catalysts in the presence of Cs2CO3. This process consists of (1) deaminative hydrogenation of secondary amides to generate primary amines and alcohols, (2) dehydrogenative coupling of the transient amines with alcohols to generate imines, and (3) hydrogenation of imines to give the formally deoxygenated secondary amine products.

A ruthenium(II)-catalyzed deoxygenative transfer hydrogenation of amides to amines using HCO2H/NEt3 as the reducing agent is reported for the first time. The catalyst system consisting of [Ru(2-methylallyl)2(COD)], 1,1,1-tris(diphenylphosphinomethyl) ethane (triphos) and Bis(trifluoromethane sulfonimide) (HNTf2) performed well for deoxygenative reduction of various secondary and tertiary amides into the corresponding amines in high yields with excellent selectivities, and exhibits high tolerance toward functional groups including those that are reduction-sensitive. The choice of hydrogen source and acid co-catalyst is critical for catalysis. - Exploring the PROTAC degron candidates: OBHSA with different side chains as novel selective estrogen receptor degraders (SERDs)
-
As the mutant estrogen receptor (ER) continues to be characterized, breast cancer is becoming increasingly difficult to cure when treated with hormone therapy. In this regard, a strategy to selectively and effectively degrade the ER might be an effective alternative to endocrine therapy for breast cancer. In a previous study, we identified a novel series of 7-oxabicyclo[2.2.1]heptene sulfonamide (OBHSA) compounds as full ER antagonists while lacking the prototypical ligand side chain that has been widely used to induce antagonism of ERα. Further crystal structure studies and phenotypic assays revealed that these compounds are selective estrogen receptor degraders (SERDs) with a new mechanism of action. However, from a drug discovery point of view, there still is room to improve the potency of these OBHSA compounds. In this study, we have developed new classes of SERDs that contain the OBHSA core structure and different side chains, e.g., basic side chains, long alkyl acid side chains, and glycerol ether side chains, to simply mimic the degrons of proteolysis targeting chimera (PROTAC) and then investigated the structure-activity relationships of these PROTAC-like hybrid compounds. These novel SERDs could effectively inhibit MCF-7 cell proliferation and demonstrated good ERα degradation efficacy. Among the SERDs, compounds 17d, 17e and 17g containing a basic side chain with a N-trifluoroethyl substituent and a para methoxyl group at the phenyl group of the sulfonamide turned out to be the best candidates for ER degraders. A further docking study of these compounds with ERα elucidates their structure-activity relationships, which provides guidance to design new PROTAC degrons targeting ER for breast cancer therapy. Lastly, easy modification of these PROTAC-like SERDs enables further fine-tuning of their pharmacokinetic properties, including oral availability.

A zirconium-based metal–organic framework, UiO-66-NH2, modified by melamine (Mlm) was used as a support for CuO nanoparticles (NPs). Melamine offered a platform for uniform and homogeneous distribution of NPs on the surface of the frameworks and made a strong bonding to the NPs to avoid undesirable leaching. UiO-66-NH2-Mlm/CuO NPs were used for the Buchwald–Hartwig C–N cross-coupling reaction to synthesize arylated anilines from phenyl iodide, bromide, and chloride and primary and secondary amines in DMF at 110°C. The catalyst was also employed for the synthesis of 2-substituted benzimidazole derivatives from various aromatic aldehydes and o-phenylenediamine in the absence of an oxidant in EtOH at room temperature. The catalyst was recyclable and reusable for several times and exhibited good stability (examined by BET, XRD, and SEM–EDX) in reaction conditions.

A rare reductive coupling of nitro compounds with organohalides has been realized. The reaction is initiated by a partial reduction of the nitro group to a nitrenoid intermediate. Therefore, not only aromatic but also aliphatic nitro compounds are efficiently transformed into monoalkylated amines, with organohalides as the alkylating agent. Given the innate reactivity of the nitrenoid, a catalyst is not required, resulting in a high tolerance for aryl halide substituents in both starting materials.

- CoII immobilized on an aminated magnetic metal-organic framework catalyzed C-N and C-S bond forming reactions: A journey for the mild and efficient synthesis of arylamines and arylsulfides

In this work, we report a simple and versatile method for the modification of a metal-organic framework (NH2-MIL53(Al)) in a step-wise manner. To characterize the synthesized nanostructured catalyst, a variety of spectroscopic and microscopic techniques including FT-IR, XRD, BET, TEM, FE-SEM, EDX, EDX-mapping, TGA, XPS, VSM, ICP-OES and CHN have been employed. Fe3O4@AMCA-MIL53(Al)-NH2-CoII NPs, which benefit from small nanocrystalline size (10-30 nm, according to the XRD and TEM data) in combination with the coexistence of magnetic nanoparticles, a metal-organic framework, and cobalt species, were found to be an excellent environment catalyst to promote the C-N and C-S cross coupling reactions. A wide range of functional substrates including electron-withdrawing and electron-donating aryl halides underwent the coupling reaction with aromatic/heteroaromatic/benzylic and aliphatic amines and sulfides. The results demonstrated that the yields of the target products were good to excellent and the catalyst can be recycled for at least seven recycling runs without a discernible decrease in its catalytic activity. Furthermore, the heterogeneity studies (such as hot filtration and poisoning tests) efficiently confirmed that the as-synthesized nanostructured catalyst is heterogeneous and completely stable under the reaction conditions. We hope that our study inspires more interest in designing novel catalysts based on using low-cost metal ions (such as cobalt) in the field of cross coupling reactions.

- Reductive Molybdenum-Catalyzed Direct Amination of Boronic Acids with Nitro Compounds

The synthesis of aromatic amines is of utmost importance in a wide range of chemical contexts. We report a direct amination of boronic acids with nitro compounds to yield (hetero)aryl amines. The novel combination of a dioxomolybdenum(VI) catalyst and triphenylphosphine as inexpensive reductant has revealed to be decisive to achieve this new C?N coupling. Our methodology has proven to be scalable, air and moisture tolerant, highly chemoselective and engages both aliphatic and aromatic nitro compounds. Moreover, this general and step-economical synthesis of aromatic secondary amines showcases orthogonality to other aromatic amine syntheses as it tolerates aryl halides and carbonyl compounds.

A palladium/copper-cocatalyzed Ullmann-type N-arylation of anilines using alkyl(aryl)sulfonium triflates as arylation reagents has been accomplished. The reaction enabled Caryl-S bond cleavage over Calkyl-S bond breakage of alkyl(aryl)sulfoniums by Pd(P(tBu)3)2/CuI and gave the corresponding N-arylated products in good to high yields. That is to say, the alkyl halide or the arylhalide and organic amine are mixed according to a certain ratio, and under light irradiation, C-N coupling reaction is carried out, so that a corresponding target product is generated for preparing the secondary amine or the tertiary amine. Compared with existing synthesis technologies, the method has the advantages that the aryl halide or the alkyl halide which is cheap and easy to get is selected as a reaction substrate, the method has good universality for different organic amine separately, moreover, the product selectivity is high, and separation and purification are easy; moreover, the synthesis method does not need to be conducted under severe conditions of high temperature and the like, a green and sustainable light source is adopted as driving force, the atom economy is high, andthe method has wide application prospects.

A simple and practical method for the denitrosation of aryl N-nitrosamines to secondary amines is reported under metal-free conditions using iodine and triethylsilane. Several reduction-susceptible functional groups such as alkene, alkyne, nitrile, nitro, aldehyde, ketone and ester were found to be very stable during the denitrosation, which is remarkable. Broad substrate scope, room temperature reactions and excellent yields are the additional features of the current methodology. (Figure presented.).
