PDK-1 inhibitors

PDK-1 (phosphoinositide-dependent kinase-1) inhibitors are compounds designed to block the activity of PDK-1, a serine/threonine protein kinase crucial for regulating signaling pathways involved in cell growth, survival, and metabolism. PDK-1 plays a central role in the PI3K (phosphatidylinositol 3-kinase)/Akt (protein kinase B) signaling pathway by phosphorylating and activating Akt, which in turn regulates downstream targets involved in cell proliferation and survival. Inhibiting PDK-1 activity disrupts this pathway, potentially impairing cancer cell growth and survival in tumors where PI3K/Akt signaling is dysregulated. PDK-1 inhibitors are under investigation as targeted therapies for cancers harboring mutations or overactivation of PI3K/Akt signaling, with the goal of improving treatment outcomes and overcoming resistance to existing therapies.
