assessments, resulted in 15 promising DORA compounds.
Identification of the best compound out of the 15 frontrunners was
[1]
finally performed by implementing a PB-PKPD model
and

assessing additional safety aspects in vitro. The efforts
culminated in the selection of compound 93, now called
daridorexant, for preclinical followed by clinical development.
Compound 93 dose-dependently promotes sleep in rats and dogs.
The sleep architecture (=relative proportions of REM vs NREM
sleep) obtained following administration of 93 is comparable to
the architecture of physiological sleep. Compound 93 is in Phase
3 clinical trials for the treatment of insomnia. Top line results of
the first pivotal phase 3 study were released on April 20th, 2020.
The study demonstrated that daridorexant improved overall sleep
and daytime performance of patients with insomnia. Results of the
second pivotal study clearly confirmed the results from the first
pivotal study.
