Reactions with cyclooctynes were found fast and highly
efficient in PBS buffer at room temperature (Figure 6). The
positive impact of the Br atom in position C4 was confirmed
by inducing an increase of the speed of reaction by one
order of magnitude (compare 9i with 10g in Figure 6) to
reach ~ 3.7 M-1.sec-1 as value of the kinetic constant k
when the cyclooctyne BCN was used. This value can be
increased to ~ 5.7 M-1.sec-1 with the use of the more
strained alkyne DBCO.
Compound 10g can be easily further functionalized
through peptide coupling. To develop a useful cleavable
linker we coupled 3-azidopropan-1-amine to 10g and
obtained compound 10h which was then easily
functionalized using Cu-catalysed click reaction with an
alkynylated coumarine substrate to get 10i demonstrating
the compatibility of the bromo-ImSyd core with the Cu-
catalyzed azide-alkyne cycloaddition reaction. Control
experiments confirmed that 10i display high reactivity
toward BCN (k = 8.142 ± 0.487 M-1.sec-1, Table S1 in SI);
this compound was also found stable even after 95 h
incubation in human plasma and reacted quantitatively
with DBCO after this period of incubation (Figure S3 in SI).

In conclusion, we reported a comprehensive study on the
chemistry of ImSyds and their reactivity towards click and
release reactions with cyclooctynes. This study allowed to
identify the most suitable substituents to increase the
reaction rate. Bromo-ImSyds were more particularly
identified as suitable reactants for click and release
reactions with cyclooctynes and were used to develop
bifunctionalized cleavable linkers with improved reactivity.
We think this biorthogonal cleavable linkers will find great
applications in chemical biology.
