Cells were first treated with different concentration of PITE for 1 h. For
control, equivalent numbers of cells were treated with respective solvents.
Subsequently, MTT (0.3 mg/mL) reagent was added to cells and incubated
for 3 h. After that DMSO was added and optical density of formazan
crystals (indicative of live cells) was measured at 570 nm.

LD imaging in Yeast cells
Yeast cells were grown till stationary phase. 1 mL of the culture was
harvested by centrifugation at 5000 rpm for 5 min and washed twice with
distilled water. The cells were resuspended in solution containing PITE
(100 L, 100 µM) and were incubated at 30 C for 30-60 minutes.
Subsequently, cells were washed twice with water and resuspended in 50
µL of water. 3-4 µL of cell suspension was used for microscopic sample
preparation. Fluorescent images were acquired by Carl Zeiss upright
fluorescence microscope using 63x oil immersion objective lens. Images
were processed by ZEN blue 2012 software. The staining with Nile Red
was performed following a reported procedure.[29b]

Isolation of LDs from yeast
Wild type BY4741 yeast cells were grown in YPD media till late exponential
phase for the isolation of LDs. Cells were converted into spheroplasts
(cells without the cell walls) by using lyticase enzyme. The spheroplasts
were lysed using homogenizer in buffer (12% W/V Ficoll, 10 mM
morpholineethanesulfonic acid, 0.2 mM EDTA, 1 mM phenylmethylsulfonyl
fluoride, pH 6.9).[29a] LDs were isolated through Ficoll density gradient

LD imaging in mammalian cells
HeLa cells were grown with or without oleic acid (optimal confluency 70-
80%) on the cover slip in 35 mm Petri dish. Cellular fixation was done using
3.7% of formaldehyde for 5 min and washed twice with PBS buffer (pH
7.4). Subsequently, the cells were stained with PITE for fluorescence
microscopic investigations. The Pearson’s correlation coefficient (for
colocalization of PITE and Nile Red) was calculated by analyzing CLSM
images using ImageJ software.[33]
