In summary, a new TYR bioluminogenic probe TYRꢀLH2 by
introducing a 3ꢀhydroxybenzyl group into Dꢀluciferin was
prepared and its property for TYR imaging in vitro and in vivo
were subsequently evaluated. The specific hydroxylation of 3ꢀ
hydroxybenzyl group by TYR combined with the clean backꢀ
ground of bioluminescence strategy endows this probe high
selectivity and sensitivity towards TYR. A detection limit of
0.11 ꢁM was obtained in a small detection volume of 100 ꢁL
which encouraged the applications of TYRꢀLH2 in vivo. Imagꢀ
ings from living cells and model animals showed that TYRꢀ
LH2 presented capability of identifying the dynamic changes
of TYR as well as discrimination of melanocytes from other
cell lines, affording new opportunities for investigation and
diagnosis of melanoma and other TYRꢀrelated diseases in
complex biological situations.
