acetylserine sulfhydrylase to alanine indicates its importance in
transimination and as a general base catalyst. Biochemistry
then eliminated as a H2O, a more efficient leaving group
than OH-. An amino acid racemase should posses some
hydrogen relay system between the catalytic residues such
as K56 and S81 of DdSR, if not, the racemization reaction
would stop after single turnover. It is thus possible that the
a-hydrogen is abstracted from D-serine by S81 and relayed
to the b-hydroxyl group of D-serine via K56. We also
cannot rule out the possibility that dehydration of D-serine
occurs after its conversion to L-serine through the
racemization.
An understanding the reason behind the inability of K56
to catalyze D-serine dehydration will help the elucidation of
the mechanism of bifunctionality of SRs catalyzing both
racemization and dehydration.
