The invention discloses a chromogenic substrate of 3-nitro-4-chlorine-benzenesulfonamide glutathione-S-transsulfurase. The chromogenic substrate is structurally characterized in that 3-nitro-4-chlorine-benzene sulfonyl chloride and one of ethanolamine, aminoethyl sulfonic acid, glycine and N,N-dimethyl-ethylenediamine are reacted to generate benzenesulfonamide; under catalysis of glutathione-S-transsulfurase (GST), the chromogenic substrate and reduced glutathione (GSH) are reacted to generate phenyl sulfide, the absorbing effect of the obtained phenyl-sulfide product is remarkably enhanced relative to the absorbing effect of the chromogenic substrate near 285 nm; the initial absorbing variation velocity near 285 nm in the process that the chromogenic substrate and the GSH are reacted under the analysis of the GST is measured, and can express the activity of the GST and be used for screening inhibitors.
