Direct synthesis of alkynes from inexpensive, abundant alcohols was achieved in high yields (greater than 40 examples, up to 95% yield) through a SO2F2-promoted dehydration and dehydrogenation process. This straightforward transformation of sp3-sp3 (C-C) bonds to sp-sp (C=C) bonds requires only inexpensive and readily available reagents (no transition metals) under mild conditions. The crude alkynes are sufficiently free of impurities to permit direct use in further transformations, as illustrated by regioselective Huisgen alkyne-azide cycloaddition reactions with PhN3 to give 1,4-substituted 1,2,3-traiazoles (16 examples, up to 92% yield) and Sonogashira couplings (10 examples, up to 77% yield).

A new series of sulfonamide based [1,2,3]-triazoles 4a-i, 5 and 6 has been prepared in 65-99% yield through Huisgen [3+2] cycloaddition reaction of 4-azido-N-substitutedbenzenesulfonamides 2a,b with various alkynes (3a-i or phenyl acetylene) in the presence of CuSO4.5H2O and sodium ascorbate in t-butanol and water (1:1) mixture at 40°C. The structures of all the newly synthesized molecules have been established on the basis of IR, 1H and 13C NMR and high resolution mass spectral analysis. Furthermore, these compounds have been evaluated for their preliminary in vitro antibacterial activity against two Gram-negative bacteria viz. Escherichia coli (MTCC 433) and Salmonella enterica ser. typhi (MTCC 733) and a Gram-positive bacterium viz. Streptococcus mutans (MTCC 497) by using MTT assay.

In this study, 20 metallocene-based compounds comprising extensive structural diversity were synthesized and evaluated as carbonic anhydrase (CA, EC 4.2.1.1) inhibitors. These compounds proved moderate to good CA inhibitors in vitro, with several compounds displaying selectivity for cancer-associated isozymes CA IX and CA XII compared to off-target CA I and CA II. Compound 6 was the most potent ferrocene-based inhibitor with Kis of 5.9 and 6.8 nM at CA IX and XII, respectively. A selection of key drug-like parameters comprising Log P, Log D, solubility, and in vitro metabolic stability and permeability were measured for two of the ferrocene-based compounds, regioisomers 1 and 5. Compounds 1 and 5 were found to have characteristics consistent with lipophilic compounds, however, our findings show that the lipophilicity of the ferrocene moiety is not well modeled by replacement with either a naphthyl or a phenyl moiety in software prediction tools.

A library of 10 novel benzenesulfonamides containing triazole-tethered phenyl 'tail' moieties were synthesized by a Cu(I) catalyzed 1,3-dipolar cycloaddition reaction (DCR) (i.e., click chemistry) between 4-azido benzenesulfonamide and a panel of variously substituted phenyl acetylenes. These compounds were very effective inhibitors (low nanomolar) of the human mitochondrial carbonic anhydrase isozymes VA and VB. Mitochondrial carbonic anhydrases are potential targets for anti-obesity therapies, acting to reduce lipogenesis through a novel mechanism of action. The inhibitors reported here should prove valuable as lead compounds to further investigate the potential of CA inhibition for this novel therapeutic application.
