The invention provides a pyridopyrimidinone compound with a structure as shown in a general formula (II) and application thereof. Researches prove that the compound provided by the invention can effectively inhibit KRAS G12C mutation. KRAS mutation accounts for a large proportion in tumors, no approved drug is obtained for treatment at present, and the compound provided by the invention has the potential to become a therapeutic drug for malignant tumors (especially non-small cell lung cancer (NSCLC) and colorectal cancer) carrying KRAS G12C mutation, and has a great application value.

A hypervalent iodine(III)-mediated intramolecular decarboxylative Heck-type reaction of 2-vinyl-phenyl oxamic acids has been developed. The unique ring-strain-enabled radical decarboxylation mechanism is preliminarily revealed. This protocol features metal-free reaction conditions and operational simplicity, allowing the lactamization of 2-vinylanilines using a readily accessible carbonyl source and the synthesis of various 2-quinolinones with excellent chemoselectivity at room temperature.

It has been found that t-butyl 2-vinylphenylcarbamate derivatives underwent iodocyclization on treatment with iodine in the presence of sodium hydrogencarbonate to afford 4-iodomethyl-1,4-dihydro-2H-3,1-benzoxazin-2-one derivatives in generally good yields. The reduction of these 4-iodomethyl derivatives with tributyltin hydride gave the corresponding 4-methyl derivatives in good yields.
