The invention relates to a preparation method of tert-butyl-1-methyl-5-oxygen subunit-1,4,9- triazaspiro [5.5] undecane-9-formyl ester, and mainly solves the technical problem that a synthetic methodsuitable for industrialization does not exist at present. The preparation method comprises the following two steps: first step, adding ethanediamine and triethylbenzylammonium chloride in dichloromethane; dropwise adding sodium hydroxide aqueous solution in reaction liquid at the temperature of 5-20 DEG C; and carrying out stirring reaction, after dissolving N-tert-butyloxycarbonyl-4-piperidone indichloromethane, dropwise adding a mixture added with chloroform in the reaction liquid, and reacting to obtain a compound 2; and second step, dissolving the compound 2 in methanol, and adding a formaldehyde aqueous solution and acetic acid; adding sodium cyanoborohydride at the temperature of 0 DEG C; and carrying out stirring reaction on the mixture, and after reaction is finished, carrying outtreatment and purification to obtain the final product. By the compound obtained in the invention, a useful intermediate or product is provided for synthesis of multiple drugs.
