The invention relates to a preparation method of 1,2-dihydrobenzo[4, 5]imidazo[1, 2-a]pyrazine-3(4H)-one derivatives. The preparation method disclosed by the invention comprises the following steps: taking a Ugi reaction as a basis, performing deprotection under acid conditions, and performing microwave-assisted ring closure to obtain benzimidazole products; performing simple after-treatment, andpreparing the 1,2-dihydrobenzo[4, 5]imidazo[1, 2-a]pyrazine-3(4H)-one derivatives having potential antitumor activities under alkaline conditions. Compared with the traditional method, the method hasthe characteristics of being readily available in raw materials, simple and short in synthetic process, simple in after-treatment, high in yield and the like.
