The invention relates to a preparation method of an asymmetric gem-diarylacetate compound, which comprises the following steps: (1) stirring and reacting a glycine derivative, a compound Ar and a catalyst aluminum trichloride in an organic solvent at 20-30 DEG C to obtain an N-aryl-[alpha]-aryl glycine derivative, wherein R is methyl or methoxyl; and (2) sequentially adding the N-aryl-[alpha]-arylglycine derivative obtained in the step (1), a compound Ar', a catalyst aluminum trichloride and a cocatalyst, and carrying out reflux reaction in an organic solvent at 70-90 DEG C to obtain the asymmetric gem-diarylacetate compound, the compound Ar and the compound Ar' are independently selected from aniline compounds, indole compounds or electron-rich arylether compounds, and the compound Ar' is different from the compound Ar. The reaction raw materials are low in price and easy to obtain, the reaction conditions are environment-friendly, the substrate application range is wide, pre-functionalization of the substrate is avoided, the operation is simple, and post-treatment is convenient.
