Synthesis of Mixed Oligonucleotides Containing 6,7-Dimethylisopterin as Modified Aglycone

6,7-Dimethyl-1-(2-deoxy-β-D-ribofuranosyl)isopterin 3 was prepared from 6,7-dimethyl-1-(2-deoxy-3,5-di-O-p-toluoyl-β-D-ribofuranosyl)lumazine via thiation and displacement of the thio function with ammonia.It was then converted into the corresponding N4-benzoyl and N4-p-nitrophenylethoxycarbonyl phosphoramidite building blocks.Mixed oligonucleotides containing 6,7-dimethylisopterin as a modified base were synthesised using solid phase phosphoramidite strategy.
