Synthesis of coumarins via PIDA/I2-mediated oxidative cyclization of substituted phenylacrylic acids

A variety of functionalized coumarins were synthesized from substituted phenylacrylic acids via PIDA/I2-mediated and irradiation-promoted oxidative carbon-oxygen bond formation. Our studies show that the oxygen in the pendant carboxylic acid group cyclizes favorably to the aryl ring that is cis to it. The main advantages of this method include good functional group tolerance and the transition-metal-free characteristic. The Royal Society of Chemistry 2013.

Distinct differentiation: β,β-Disubstituted acrylic acids with sterically similar geminal diaryl groups can be hydrogenated with excellent enantioselectivities in the presence of a RhI complex formed in situ with two-component ligands, a chiral secondary phosphine oxide (SPO) and an achiral phosphine (Ph3P). The utility of arenediazonium salts immobilized in imidazolium-ILs [BMIMPF6 and BMIMBF4] for facile, high yielding, synthesis of olefins via the Pd(OAc)2-catalyzed Matsuda-Heck arylation reaction has been demonstrated. The reaction can also be performed as a two-step process in the IL starting from ArNH2, by sequential in-situ diazotization-arylation. Simple product isolation and the recycling/re-use of the IL are additional advantages of this one-pot method.

Using a known dual PPARα/γ activator (5) as a structural template, SAR evaluations led to the identification of triple PPARα/γ/δ activators (18-20) with equal potency and efficacy on all three receptors. These compounds could become useful tools for studying the combined biological effects of PPARα/γ/δ activation.

Ethyl cinnamate reacts with several para-substituted aryl iodides under Jeffery-Larock conditions (Pd(OAc)2, NaHCO3, n-Bu4NBr, DMF, Δ) to give ethyl (E)-3-aryl-3-phenylpropenoates as major compounds. The reaction of para-substituted ethyl cinnamates with iodobenzene under analogous conditions affords the corresponding Z isomers. The initially stereodefined alkene formed under Heck conditions undergoes a slow isomerization.
