We have developed an alternative method for the synthesis of 2'-modified 4'-thionucleosides. The fusion of 3,5-di-O-benzoyl-1-bromo-2-deoxy-2-fluoro- 4-thio-α-D-arabinofuranose, prepared from 1,2:5,6-diisopropylidene-α-D- allofuranose, with persilylated N4-acetylcytosine predominantly gave a β- anomer of protected 4'-thionucleoside, which was then deprotected to give 4'- thioFAC.
