The invention provides a preparation method of 2-methoxy-5-acetamido-N, N-diacetoxyethyl aniline. The method comprises the following steps: (1) carrying out hydroxylation reaction on o-methoxy m-acetamido aniline and ethylene oxide to obtain a hydroxylate 2-methoxy-5-acetamido-N, N-dihydroxyethyl aniline; and step 2), carrying out an esterification reaction on the hydroxylate 2-methoxy-5-acetamido-N, N-dihydroxyethyl aniline and acetic anhydride and/or acetic acid, so as to obtain an esterified compound. The 2-methoxy-5-acetamido-N, N-diacetoxyethyl aniline prepared by the method disclosed by the invention is low in cost and simple in steps, and the obtained product is high in yield, high in purity and more suitable for workshop production.
