Acetylation of 2-amino-4-(4-methoxyphenyl)-6-oxo-1,6-dihydropyrimidine-5-carbonitrile (2) using acetic anhydride afforded N-pyrimidinylacetamide derivative 3 which upon formylation using Vilsmeier-Haack reagent produced β-chloroenaldehyde derivative 4. The chemical?behavior of compound 4 was investigated toward some primary amines as well as heterocyclic amines yielding the corresponding Schiff bases. Compound 4 is used as a building block for the construction of nitrogen heterocyclic compounds such as pyrazoles,?pyrimidines, pyridopyrimidine and diazepine through its reaction with a diversity of bifunctional nucleophiles such as hydrazines, guanidines, 6-aminouracil and ethylenediamine. The biological activity of the synthesized compounds was investigated. Structures of the newly synthesized products were deduced on the basis of their analytical and spectral data.

In this work, we describe a multicomponent microwave-mediated synthesis of 10 2-amino-6-aryl-4-oxo-1,6-dihydro-pyrimidine-5-carbonitriles in good chemical yields (44-67%), four of them not related in earlier literature. All pyrimidinones synthesized herein had their antimicrobial activity evaluated against the following microorganisms: Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. Two of the synthesized substances displayed good antimicrobial activity against P. aeruginosa and S. aureus, two bacteria responsible for nosocomial infections. Synthesis of some new cyanopyrimido[1,2-a]pyrimidinones 5-22 have been achieved via interaction of 2-amino-6-anisyl-5-cyano-4(3H)-pyrimidinone (1) with some heterocycles having a vicinal chloroester, chlorocyano or mercaptocyano group, dimethyl acetylenedicarboxylate, active methylene compounds, ethyl 2-acetyl-3-anisylpropenoate, ethyl 3-aryl-3- cyanopropenoates, ethyl 2-cyano-3-ethoxyacrylate and some enones or enals. Some of the isolated products were subjected to biological screening tests.
