The invention discloses a synthesis method of 4-methoxycarbonyl-2-methylthiophene-3-sulfonyl chloride, which comprises the following steps: S1) reacting methyl 5-methyl-4-oxotetrahydrothiophene-3-formate in a formula (I) with methylsulfonyl chloride under the action of a catalyst to obtain an intermediate in a formula (II); S2) mixing the intermediate shown in the formula (II), anhydrous sodium sulfide and a polar aprotic solvent for reaction, introducing oxygen, and performing aromatization to obtain an intermediate reaction solution shown in a formula (III); S3), sequentially carrying out chlorine oxidation and conventional post-treatment on the intermediate reaction liquid in the formula (III) to obtain a target compound that is 4-methoxycarbonyl-2-methylthiophene-3-sulfonyl chloride ina formula (IV). The method has the advantages of short steps, simple operation, high yield, low cost and less wastewater, and is more suitable for industrial production.

The invention relates to the field of organic synthesis, in particular to a synthetic method for 4-methoxycarbonyl-2-thiotolene-3-sulfonyl chloride. The synthetic method for the 4-methoxycarbonyl-2-thiotolene-3-sulfonyl chloride comprises the following steps: carrying out diazotization reaction in a micro-channel reactor by taking a 4-methoxycarbonyl-2-thiotolene-3-amino hydrochloric acid solutionand a sodium nitrite aqueous solution as raw materials to synthesize diazonium salt; and carrying out chlorosulfonation on the obtained diazonium salt and sulfur dioxide gas under the effect of a catalyst and a surfactant in the micro-channel reactor to obtain the 4-methoxycarbonyl-2-thiotolene-3-sulfonyl chloride. The synthetic method for the 4-methoxycarbonyl-2-thiotolene-3-sulfonyl chloride has the advantages that the micro-channel continuous flow reactor is adopted, the reaction time is shortened to few tens of second to several minutes from traditional several hours and even tens of hours, and the reaction efficiency is improved remarkably; because two phases of the raw materials are uniformly mixed in a micro-channel, the temperature is accurately controlled, and the conversion rate of the raw materials and the selectivity of the product are improved obviously; and meanwhile, the whole feeding and mixing and reaction process is a continuous-flow reaction process, and influencesof air to reaction in conventional intermittent reaction are avoided.

The method comprises the steps: reacting 2-methyl-4-methoxycarbonyl thiophene, sulphur and anhydrous tetrahydrofuran with a lithium diisopropylamide tetrahydrofuran solution and then dropping into a dichloromethane solution which chlorine gas is dissolved in. The method for preparing the 2-methyl-3-thiophene-5-methoxycarbonyl sulfonyl chloride from the chlorine gas disclosed by the invention has the advantages of alittle industrial wastewater and high reaction yield.
