A safe and efficient route for the preparation of nitro-, dinitro-, and trinitrophenyl chloroformates

A safe and simple method has been developed for synthesis of chloroformates derived from 2-nitrophenol, 4-nitrophenol, 2,4-dinitrophenol, 2,4,6-trinitrophenol, and pentafluorophenol, the yields being in the range of 72-96%. The method is based on a heterogeneous reaction of the substituted phenol with phosgene dissolved in dichloromethane catalyzed with solid anhydrous potassium carbonate. The synthesis of 2,4,6-trinitrophenyl chloroformate takes place only in the presence of a phase-transfer catalyst, in this case the easily recovered α,ω-dimethoxy poly(ethylene glycol) (PEG; M = 2kDa). The only waste products of the suggested synthesis of the mentioned esters are sodium chloride and potassium chloride. The method markedly reduces the explosion risk connected with drying and handling of salts of mono-, di-, and trinitrophenols and is much more environmentally friendly than the currently used methods. In order to achieve site specific delivery of NO, we designed conjugates of cephalosporin with NO donors. NO donors such as cupferron and SIN-1 were evaluated as potential choices for conjugates. Cephalosporin conjugated with SIN-1 demonstrated promising beta-lactamase dependent NO releasing ability.

An investigation of methods to efficiently prepare activated N-methylcarbamates is reported. N-(Methylcarbamoyloxy)succinimide (3a), aryl N-methylcarbamates 3b-d and 2,2,2-trifluoro-1-(trifluoromethyl)ethyl N-methylcarbamate (3e) have been prepared in 70-80% yields from the corresponding chloroformates 5a-e, which were prepared as crystalline solids by the condensation of trichloromethylchloroformate (1) or bis(trichloromethyl)carbonate (2) with hydroxy compounds 4a-e in high yields.

The reaction of pentafluorophenol, 2,2,3,3-tetrafluoropropyl alcohol, and 2,2,3,3,4,4,5,5-octafluoropentyl alcohol with carbon tetrachloride in the presence of aluminum chloride were investigated.The main products from the reaction of pentafluorophenol with these reagents are pentafluorophenoxytrichloromethane, di(pentafluorophenol) carbonate, and tri(pentafluorophenoxy)chloromethane.The ratios of the products depend on the reaction conditions.The use of a large excess of carbon tetrachloride leads to the preferential formation of pentafluorophenoxytrichloromethane.The addition of water promotes the formation di(pentafluorophenyl) carbonate.When heated with carbon tetrachloride and aluminum chloride and subsequently treated with water, 2,2,3,3-tetrafluoropropyl and 2,2,3,3,4,4,5,5-octafluoropentyl alcohols are converted into di(2,2,3,3-tetrafluoropropyl) carbonate and di(2,2,3,3,4,4,5,5-octafluoropentyl) carbonate respectively.

Compounds useful as inhibitors of the enzyme elastase are of the following general formula: STR1 wherein Z is selected from the group consisting of R"O--Suc--where R" is lower alkyl of 1 to 3 carbon atoms and CF 3 CO--; X is oxygen or sulfur; R' is selected from the group consisting of straight or secondary branch-chained alkyl of 1 to 4 carbon atoms, alkenyl of 2 to 3 carbon atoms, alkynyl of 2 to 4 carbon atoms, cycloalkyl of 3 to 6 carbon atoms, and benzyl, and R is selected from the group consisting of susbstituted or unsubstituted phenyl wherein the substituents are selected from the group consisting of nitro, and pentafluoro; benzyl, CH 2 CF 2 CF 2 CF 3, 1-lower alkyl tetrazolyl, 1-phenyltetrazolyl, 2-thioxo-3-thiazolidinyl-, pyridyl and benzothiazolyl, provided that when R is paranitrophenyl, R' is other than tertiary-butyl, benzyl or cyclohexyl, and when X is sulfur, R is other than benzyl. "
