Radical substitution with azide: TMSN3-PhI(OAc)2 as a substitute of in3

TMSN3 and PhI(OAc)2 were found to promote high-yield azide substitution of ethers, aldehydes and benzal acetals. The reaction is fast and occurs at zero to ambient temperature in acetonitrile. However, it is essential for the reaction that TMSN3 is added subsequent to the mixture of PhI(OAc)2 and the substrate. A primary deuterium kinetic isotope effect was found for the azidonation of benzyl ethers both with TMSN3-PhI(OAc)2 and with IN3. Also a Hammett free energy relationship study of this reaction showed good correlation with σ+ constants giving with ρ-values of -0.47 for TMSN 3-PhI(OAc)2 and -0.39 for IN3. A series of saccharidyl-triazoles structurally related to acarbose were prepared and tested as inhibitors of glycosidases. They share in common a 1,4,5-trisubstituted 1,2,3-triazole heterocycle as a functional element able to interact with the active site of the target enzymes. First, it was established that the heterocyclic core exhibits a moderate but highly selective α-glucosidase inhibitory activity. Then, it was confirmed that the inhibitory properties could be modulated by conjugation from one to five carbohydrate residues. The present study includes the regio- and stereocontrolled synthesis of novel non-fused 1,2,3-triazolo- pseudooligosaccharides as well as their evaluation as new glycosidase inhibitors.

A novel photoluminescence lifetime probe (Ir-TB) has been developed for the detection and imaging of hCE2 in living cells. A large lifetime increase by around 300 ns after the enzymatic reaction makes it an ideal tool to distinguish hCE2-hydrolyzed probes from those non-hydrolyzed ones via PLIM for the first time.

A series of N6-adenosine derivatives were synthesized by alkylation of N6-acetyl-2',3',5'-tri-O-acetyladenosine (1) with alkyl halides and alcohols. It was shown that propargyl derivative 2a is a good substrate for copper(I) catalyzed Huisgen [3+2] cycloaddition with azides. This click-reaction can be used for preparation of the libraries of 1,2,3-triazolyl modified adenosines. Biological activities of N6-adenosines were studied in two plant and six human cancer cell assays. The remarkable parallel between cytokinin and cytotoxic activities was found. The most cytokinin active compounds 3c-3e at the same time appeared to be the most potent cytotoxic agents.
