The invention provides a synthesis method of deuterated hexamethylphosphoramide-D18, which comprises the following steps of in the presence of triethylamine, carrying out substitution reaction on deuterated dimethylamine hydrochloride and phosphorus oxychloride to obtain the deuterated hexamethylphosphoramide-D18. In the reaction, a reaction solvent is dichloromethane, the reaction temperature isroom temperature, and the reaction time is 16 hours. The method has the following technical effects that deuterated dimethylamine hydrochloride sold in the market is used for replacing deuterated dimethylamine gas for the first time and reacts with phosphorus oxychloride to prepare the deuterated hexamethylphosphoramide-D18.
