The invention relates to a preparation method of cresol purple perchlorate and belongs to the field of organic synthesis. The preparation method comprises the following steps: dropwise adding acetic anhydride into an m-aminophenol solution, and maintaining the temperature at 0-5 DEG C for reaction; then adding water and sodium nitrite, dropwise adding phosphoric acid, maintaining the temperature, and carrying out centrifugal separation, and washing, so as to obtain 2-nitroso-5-acetaminophen; and adding an alcohol solvent, dropwise adding perchloric acid, carrying out reflux reaction at a room temperature, cooling, filtering, and drying, so as to obtain cresol purple perchlorate. Compared with a literature synthesis method, the preparation method has the advantages that the reaction steps are simplified, the operation is easy, the product quality is high, and the preparation method has extremely strong process competitive advantages.
