An efficient one-pot synthesis of hydroxynaphthyl-substituted glycine derivatives from 2- or 1-naphthol, glyoxylic acid, and benzyl carbamate via a modified Mannich reaction is described. The enantiomers were successfully separated on analytical and semipreparative HPLC columns. Their absolute configurations were determined by CD analysis supported by TDDFT CD calculations. A facile preparation of novel α-hydroxy-α-(2-hydroxy-1- naphthyl)acetic acid by a base-catalyzed condensation of 2-naphthol with glyoxylic acid is described. One of its derivatives, α-methoxy-α-(2- methoxy-1-naphthyl)acetic acid, has successfully been resolved.
