A photocatalyst- and transition-metal-free visible-light-induced cyclization of ortho-halothiobenzanilides has been developed. Upon irradiation with visible light, substrates undergo dehalogenative cyclization to 2-aryl benzothiazoles with high efficiency and selectivity. This photocyclization exhibits a high tolerance to various functional groups, is applicable for the synthesis of 2-alkyl benzothiazoles, and is easy to set up for gram-scale reaction.

N-(2-bromophenyl) alkyl thioamide such as an N-(2-bromophenyl) alkyl thioamide derivative is used as a raw material, sodium phosphate is used as alkali, the biphenyl benzothiazole compound is prepared through a one-pot method, the reaction is carried out under visible light irradiation, metal ions disclosed in the prior art are not needed, and the reaction yield is high.

An efficacious and mild KOtBu-promoted intramolecular C-S cross-coupling of ortho-iodothioanilides in conjunction with a catalytic quantity of phenanthroline as an additive has been described for the convenient synthesis of 2-substituted benzothiazoles. The methodology is suitable for attaining a wide variety of 2-alkyl- and 2-aryl-substituted benzothiazoles. Single-crystal XRD, DFT calculations, NMR, and UV studies suggest that halogen bonds between the units of ortho-iodothioanilides may assist in the electron transfer process.

A two-step protocol involving Pd-catalyzed cyclization of readily accessible p-bromophenyl-aldimines to 2-bromophenyl-benzoxazole/benzothiazole in [BMIM][PF6] or [BMIM][BF4] as solvent, followed by the Suzuki, Heck, and Sonogashira cross-coupling reactions catalyzed by Pd or Ni is described that generates libraries of diversely substituted 2-aryl-/heteroaryl-benzoxazoles/benzothiazoles in respectable isolated yields under mild reaction conditions. The feasibility to perform the two-steps in sequence in one-pot starting from the aldimines is also demonstrated and the potential for recycling/reuse of the IL solvent is also shown.

An iodine-catalyzed aerobic dehydro-aromatization has been developed, providing straightforward and efficient access to various benzoazoles and benzoazines. The present transition-metal-free protocol enables the dehydro-aromatization of tetrahydrobenzazoles and tetrahydroquinolines with molecular oxygen as the green oxidant, along with some other N-heterocycles. Hence, a broad range of heteroaromatic compounds are generated in moderate to good yields under facile reaction conditions.

Herein, we report the facile synthesis of various benzimidazoles and benzothiazoles by using the NH2-MIL-125(Ti) MOF as an efficient oxidant-free heterogeneous catalyst with good yield. Adsorption of the substrate on the NH2-MIL-125(Ti) MOF surface through electron deficient Ti4+ sites initiates the reaction. The broad substrate scope and high reusability of this catalyst are attractive for synthesis of a wide range of medicinally active benzimidazole and benzothiazole derivatives.

A ZnCl 2 -catalyzed regio- and diastereoselective one-pot sequential Knoevenagel condensation/1,3-dipolar cycloaddition reaction of CH acids (oxindole, 1,3-indandione, and 3 H -pyrazol-3-one), aldehydes, and dibromoformaldoxime has been established. The method allows the synthesis of diversely functionalized spiroisoxazolines in good isolated yields under mild reaction conditions. Moreover, the preparation of spiroindene-isoxazole-1,3-diones containing benzoimidazole or benzothiazole moieties as new ligands for the metal-catalyzed coupling reactions and C-H activation is demonstrated.

A S 8 -mediated directed decarboxylative redox-cyclization strategy for the synthesis of 2-substituted benzothiazoles from o -iodoanilines, arylacetic acids, and elemental sulfur catalyzed by cheap copper metal has been developed. This reaction is operationally simple, ligand-free, compatible with a wide range of functional groups, and provides the desired products in good to excellent yields. In addition, a gram-scale experiment was carried out to furnish PMX 610, an antitumor drug.

Selective construction of 2-substituted benzothiazoles from o-iodoaniline derivatives S8 and N-tosylhydrazone via a copper-promoted [3 + 1 + 1]-type cyclization reaction has been realized. In the protocol, the carbon atom on N-tosylhydrazone could be regulated to construct benzothiazole by changing the reaction system. Furthermore, the transformation has achieved the construction of multiple carbon-heteroatom bonds.
