Reaction of 3-amino-2-alkoxycarbonylthiophens with compounds such as diethyl malonate or ethyl cyanoacetate produced amides, which were cyclised to 6-substituted-7-hydroxythienopyridin-5(4H)-ones by treatment with sodium hydride in dimethyl-formamide.Hydrolysis of the 6-ethoxycarbonyl derivative yielded 7-hydroxythienopyridin-5(4H)-one; some aspects of the chemistry of this substance, and of chloro-compounds derived from it, have been studied.
