The present invention discloses a salfaprodil impurity A and a preparation method thereof, and belongs to the field of medicine and chemical industry, wherein the structure of the salfaprodil impurity A is represented by a formula (VIII). According to the preparation method, 2,3,5,6-tetrafluoro-4-methyl benzoic acid is adopted as a raw material, and a seven-step reaction is performed to obtain the salfaprodil impurity A. With the synthesis of the salfaprodil impurity A, the qualified reference substance is provided for salfaprodil quality control. The formula (VIII) is defined in the instruction.

Nitromethane and nitroethane in the presence of DBU attack preferentially the para position of pentafluorobenzonitrile and methyl pentafluorobenzoate to give the addition products in good yield. The resulting nitro compounds can be converted to tetrafluorotoluene or ethyl benzene derivatives by tri-n-butyltin hydride. TiCl3 solution neatly transforms the nitroethyl compounds into corresponding ketones. With a base, the nitro compounds can be used to extend chain length using Michael acceptors like ethyl acrylate and acrylonitrile.
