Sulfoxonium ylides as carbene precursors couple smoothly with thioureas in the presence of 5 mol% of rhodium(II) acetate dimmer via carbenoid insertion to afford the corresponding 2-aminothiazoles with high chemoselectivity, providing a facile and efficient approach to access a variety of 2-aminothiazole derivatives with good functional groups tolerance.

The invention relates to a novel green synthesis method for efficiently synthesizing aminothiazole derivatives through transition metal catalyzed carbene insertion/cyclization reaction. According to the method, sulfur ylide serves as a carbene donor, carbene is catalyzed by a transition metal to be subjected to an insertion/cyclization reaction, a C-S bond is efficiently formed, and the 2-aminothiazole derivative is constructed. Compared with the traditional method, the method has the advantages that the raw materials are easy to obtain, the steps are simple, the mild sulfur ylide reagent is used for replacing a halogenating reagent required in the traditional synthesis method, and the method is a mild, quick, simple, convenient, effective and environment-friendly method for preparing the2-aminothiazole and the derivative thereof and has a wide application prospect.
CuBr2 mediated synthesis of 2-Aminothiazoles from dithiocarbamic acid salts and ketones

In a one-pot procedure, CuBr2 has been used as a efficient desulfurizing agent in the synthesis of 2-aminothiazoles by the condensation of in situ-generated 1-substituted thioureas from their dithiocarbamic acid salts, with in situ-generated α-bromoketones from ketones. All reactions were carried out under optimized reaction conditions and gave the target products in 61–95% yield.

Several straightforward and practical processes have been established for the construction of 2-aminothiazoles, 1,3-thiazoles and 1,3-selenazoles from aryliodoazides. These strategies successfully proceed with a wide spectrum of substituted thioamides and its derivatives producing the resulting five-membered heterocycles obtained in satisfactory yields. The unique features of these protocols are operational simplicity and highly functional group tolerance, which make them convenient and practical routes for the preparation of various libraries of 2-aminothiazoles, 1,3-thiazoles, and 1,3-selenazoles.

The heterogeneous coupling reaction of oxime acetates with isothiocyanates was achieved at 110°C in toluene in air in the presence of a bidentate nitrogen-functionalised MCM-41-immobilised copper(I) complex (MCM-41-2N-CuI) with Cs2CO3 as base to afford a variety of 2-aminothiazoles in good yields. The MCM-41-2N-CuI catalyst can be easily recovered by a simple filtration and reused at least eight times without significant loss of activity.

A new strategy for 2-aminothiazoles is developed via the copper-catalyzed coupling of oxime acetates with isothiocyanates. Various 4-substituted and 4,5-disubstituted 2-aminothiazoles were formed smoothly under mild reaction conditions. This process involved copper-catalyzed N-O bond cleavage, activation of vinyl sp2 C-H bonds, and C-S/C-N bond formations. It is noteworthy that the oxime acetates were used not only as a substrate but also as a single oxidant.

The first ever tandem one-pot synthetic protocol for the synthesis of thiazoles/selenazoles from alkynes via the formation of 2,2-dibromo-1- phenylethanone is reported. The reaction is catalyzed by β-cyclodextrin in aqueous medium and resulted in good yields.
